Objective:To study the effects and mechanism of Apium graveolens L.root in Xinjiang and main components on lipid metabolism in L02 cells and HepG2 cells in vitro fat accumulation model.Methods:1?A model of hepatic steatosis in the L02 cells and HepG2 cells was induced by free fatty acids?FFA?,and the in vitro fat accumulation model of the two cells was established;The oil red O staining and biochemical parameters were used to verify whether the model was successfully established;The toxicity of the drug to the cells was determined by CCK-8 kits,and the high,medium and low concentrations in the safe range of the drug were screened.2?This experiment set blank?Con?group,model?FFA?group,fenofibrate?FF?group,apigenin?AGN?group,apiin?AIN?group,extract of Apium graveolens L.root?AGRE?group.The content of adipokines and inflammatory factors was determined by enzyme-linked immunosorbent assay?Elisa?;The expressions of ADPN mRNA,AMPK mRNA and SREBP-1c mRNA were detected by qRT-PCR;P-AMPK?,P-ACC,ACC and SREBP-1C proteins were detected by Western-blot technique.Results:1?A model of hepatic steatosis in the L02cells and HepG2 cells was successfully established.The IC500 of AGN,AIN,3?-methoxy apiin?3?-MI?and AGRE on L02 cells were 49.90,541.94,224.36,896.00?g/mL,respectively;The IC500 of HepG2 cells was 64.52,430.23,302.68,and 961.00?g/mL.The levels of TG and TC in the cells were effectively reduced by AGN,AIN and AGRE?P<0.05?.2?AGRE,AGN,AIN and FF significantly decreased the levels of IL-6 and TNF-?compared with the FFA group,?P<0.05?,and the expression of adiponectin?ADPN?secreted protein in FF group increased significantly.AGN group,AIN group and AGRE group increased the expression of ADPN mRNA and AMPK mRNA in a dose-dependent manner,and decreased the expression of SREBP-1c mRNA.In addition,the expression of P-AMPK?and P-ACC protein was also significantly increased,and the expression of SREBP-1C and ACC protein was decreased.Conclusion:AGRE has a reliable lipid-lowering and liver-protecting effect.Its mechanism may be through the regulation of AMPK/SREBP-1c signaling pathway in lipid metabolism.This study can provide experimental evidence for the development of drugs for the prevention and treatment of diseases related to lipid metabolism disorders such as non-alcohol fatty liver disease?NAFLD?.
