Objective:We aimed to study the mechanism of action of Yi Mai Ke Li(YMKL)in resistance to atherosclerosis by establishing rabbits models to perform experiments and collected clinical cases to observe the variations in intima-media thickness of patients.Methods:1.Experiment Research: 24 new zealand rabbits were randomly divided into three groups,and there were 8 rabbits in every group.The 1st group were fed a high cholesterol diet;the 2nd group was given a high cholesterol diet with YMKL(1g·d-1·Kg-1);the third one to a regular diet.After 4 months,we took a bit of blood from the ear veins and applied enzyme methods to measure every rabbit's blood lipid level.We took chest aortas after executing rabbits and used HE staining to observe atherosclerosis.At the same time,we used immunohistochemical staining to observe lox-1 protein expressions and used RT-PCR to investigate gene expressions.2.Clinical research:We divided 60 cases randomly into two groups:the treatment group of “Yi Mai Ke Li”and ordinary western medicine therapy and the control group of ordinary western medicine cure.The treatment period last for 6 months.We observed the variations of clinical signs,symptoms and assay indexes before and after treatment.Results:1.Experiment research reveals that:(1)Contrast of lipids in three groups:The degree of blood lipid level was obviously lower in YMKL group than that in hyperlipemia group.(2)The degree of atherosclerosis of 3 groups in comparison:The intima thickness and intima-media thickness ratio(I/M thickness)of YMKL group were prominently decreased than the high-cholesterol group(P<0.05).(3)LOX-1 protein expression of 3 groups: the YMKL group decreased clearly contrasted with the high-cholesterol group,(P<0.05),and the natural group without expression.(4)Three groups of lox-1 gene expression:YMKL group obviously declined in comparison with the high-cholesterol group,.2.Clinical research:After six months,we concluded that,contrasted with the control group,total-effective rate of treatment group was 86.67%,the difference is predominant(P<0.05).We found the IMT was obviously lower after treatment.Conclusions:1.YMKL can significantly inhibite the protein expression and gene expression of lox-1 in AS,and protect the function of endothelial cells.2.YMKL can apparently decrease the thickness of IMT.3.YMKL is effective and safe for atherosclerosis.
