Purpose:1.To investigate the selection of acupuncture points in the treatment of insomnia symptoms with electroacupuncture single point and compatibility acupoints;2.To use EEG monitoring to assess the success of the insomnia rat model;3.To discuss the sleep relatedness of electroacupuncture intervention in cerebrospinal fluid of insomnia rats The mechanism of neurotransmitter regulation is to study the therapeutic effect of electroacupuncture with single acupoints and compatibility with acupoints on insomnia,and provide scientific theoretical basis for experimental animal studies of neurobiological mechanisms of insomnia and clinical acupuncture treatment of insomnia.Method:A total of 120 experimental male SD rats were allocated according to a randomized method.There were 60 rats in the electrophysiological group,10 in each group,and 60 non-electrophysiological groups,10 in each group.The electrophysiological group and the non-electrophysiological group were the same group of 6 groups,namely normal group,model group,Shenmen group,Baihui group,Sanyinjiao group and Shenmen-baihui-Sanyinjiao group,among which Shenmen-hundred The Association-Sanyinjiao Group is also called the"San Cai Cave Group."(1)The electrophysiology group had to carry out electrode embedding surgery before the modeling.After the rats were placed in a self-made plexiglass cage,they were fed for a week and then molded.(2)The model of insomnia rat was duplicated by intraperitoneal injection of p-chlorophenylalanine(PCPA)once/1d for 2 days.After rats in the first intraperitoneal injection of 28?30h,the circadian rhythm of rats disappeared,and the daytime activities were significantly different from the blank control group,that is,the model was successfully reproduced.(3)The treatment was started on the third day after the model was established.The material was taken after the last treatment.The ultrasonographic liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS)target detection was used to analyze the neurotransmitter histamine and trough in the cerebrospinal fluid of experimental rats.Concentrations of 5-hydroxyindoleacetic acid,dopamine,serotonin,and its metabolite.Result:(1)PSG analysis showed that the awakening time of the model group rats on the 4th day was significantly prolonged(p<0.05)compared with the normal group rats.Compared with the model group,the awakening time of the treatment group rats was significantly reduced.(p<0.05),and the awakening time of the combined group was significantly less than the other three treatment groups(p<0.05).(2)The results showed that the final concentrations of histamine,glutamate,and dopamine in the cerebrospinal fluid of the model group were significantly higher than those in the normal group(p<0.05),and the final concentrations of 5-hydroxytryptamine and 5-hydroxyindoleacetic acid were significant.Lower than the normal group(p<0.05);compared with the model group,acupuncture at Shenmen points significantly reduced the concentration of histamine,glutamic acid,and dopamine in the cerebrospinal fluid(p<0.05),and acupuncture at Baihui reduced the valley.The concentration of glutamic acid and dopamine(p<0.05),the concentration of glutamic acid and dopamine after acupuncture at Sanyinjiao points were significantly reduced(p<0.05),and the concentration of glutamate and histamine after acupuncture and acupoint combination(acupoint compatibility)were significantly reduced.All significantly decreased(p<0.05),and histamine and dopamine concentrations were significantly lower than Shenmen,Baihui and Sanyinjiao groups(p<0.05);acupuncture at Shenmen,Baihui,Sanyinjiao and Hexue were Significantly increased the concentration of 5-HT and 5-hydroxyindoleacetic acid in cerebrospinal fluid(p<0.05),and the effect of acupoints was significantly higher than that of the three single acupoint groups(p<0.05).Conclusion:1.Electroencephalography can be used as an indicator to evaluate the exact validity of rat insomnia model replication.2.From the aspect of neurotransmitters and electrophysiology,it can be found that electro-acupuncture can effectively regulate the symptoms of insomnia in rats.3.Electroacupuncture can effectively reduce the content of glutamate,dopamine and histamine in the cerebrospinal fluid of rats,and reduce the content of 5-HT and 5-HIAA in the excitatory neurotransmitters,indicating that compatibility of acupoints can regulate insomnia Better than a single point,and the compatibility of acupoints is more effective and plays a role in tranquilizing the mind and regulating the balance of yin and yang.
