Objective:To evaluate the plasma level changes of 8-hydroxydeoxyguanosine(8-OHd G),Malondialdehyde(MDA)and DJ-1 protein in the early diagnosis and the Relation with the Severity in in Patients with Parkinson's Disease.Methods:40 Patients with idiopathic Parkinson's disease were recruited from December 2016 to June 2017 in the department of Neurology,Fujian Provincial Hospital.Exclude criteria:dementia or has a history of severe mental illness,and severe heart?brain?liver? kidney?blood?endocrine metabolic disease and tumor,progressive paralysis on nuclear,multiple system atrophy and basal ganglia cortex degeneration superposition of Parkinson's syndrome.40 health individuals were recruited as control group.The paramiters including the age,genda,duration,Hoehn&Yahr(H-Y)scales,and the unified Parkinson's disease rating scale part ? scores were recorded.According to the severity of the disease,patients were divided into early stage PD group with H-Y scales of 1-2 degree and advanced stage PD group with H-Y scales more than 2 degree.Plasma Malondialdehyde(MDA)level was tested by using thiobarbituric acid assay,and plasma 8-hydroxy-2-deoxyguanosine(8-OHd G)and DJ-1 protein was tested by using enzyme linked immunosorbent assay.Results:Plasma MDA level in PD group was significantly higher than that in control group(P=0.004,P<0.05).Plasma 8-OHd G level in PD group was significantly higher than that in control group,(P=0.027,P<0.05).There was no significantly statistical difference in Plasma DJ-1 level between control group and PD group,(P=0.093,P>0.05).There was significantly statistical difference in Plasma MDA level between control group,early stage PD group,and advanced stage PD group,(P=0.011,P<0.05).Compared with the control group,early stage PD group and advanced stage PD group had a higher level of Plasma MDA(respectively P=0.045?P=0.005).There was significantly statistical difference in Plasma 8-OHd G level between control group,early stage PD group,and advanced stage PD group(P=0.048,P<0.05).Compared with the control group,early stage PD group had a higher level of Plasma 8-OH-d G,P=0.015(P<0.05).No significant difference in plasma MDA,8-ohdg and dj-1 levels in different H-Y stages.Conclusion:Plasma level of MDA and 8-OHd G,but not DJ-1 protein,are biomarkers of the oxidative stress status in PD and are used as a supplementary method for diagnosis and evaluation the severity of disease for PD.Excessive abnormal oxidative stress injury plays an important role in the pathogenesis of PD,anti-oxidative treatment should be highlighted in PD patients.
