Objective Explore the abnormal methylation status of SLC5A8 gene in thyroid carcinoma,paracancerous tissues,and normal thyroid tissues,and to detect the expression of SLC5A8 gene in various tissues,and to synthesize the mechanism of SLC5A8 gene in thyroid carcinoma analysis.Methods Thirty-five patients with thyroid carcinoma hospitalized in the basic surgical department of the Affiliated Hospital of Hebei University from January 2017 to December 2017 were selected,including 26 cases of papillary thyroid carcinoma,accounting for 74.29%,and 7 cases of thyroid follicular carcinoma,accounting for 20%.Medullary thyroid carcinoma in 2 cases,accounting for 5.71%.The thyroid carcinoma tissues,paracancerous tissues,and marginal tissues were collected,methylation-specific PCR(MSP)was used to detect and compare the methylation status of SLC5A8 gene in carcinoma tissues,paracancerous tissues,and marginal tissues.Methylation status of SLC5A8 gene in each tissue,and further using real-time fluorescence quantitative PCR(qRT-PCR)detection of SLC5A8 gene mRNA expression in each tissue.Results The methylation positive rate of SLC5A8 gene was analyzed and statistically analyzed in the general conditions among various groups,suggesting age,gender,tumor type and lymph node metastasis,and there was no significant difference(P>0.05).The SLC5A8 gene methylation PCR results showed that 35 cases,the carcinoma tissue samples were found after the methylation amplification band as 22 cases,the ratio was 62.86%;cancerous tissue methylation in 4 cases,accounting for 11.43%;Methylation of marginal tissue occurred in one case,accounting for 2.86%.The positive rate of methylation of SLC5A8 gene was significantly higher in thyroid carcinoma tissues and paracancerous tissues combined with marginal tissue(p<0.01).Further comparisons showed that the positive rate of SLC5A8 gene methylation in carcinoma tissues was positive.Compared with paracancerous tissues and marginal tissues,the difference was statistically significant(P<0.01);the results of methylation of SLC5A8 gene in paracancerous tissues and marginal tissues were positive(P>0.05).Further qRT-PCR method was used to detect the expression of SLC5A8 gene mRNA in each tissue.The analysis showed that there was a statistically significant difference between the carcinoma tissue,paracancerous tissues and marginal tissues.Further analysis showed that there was a statistically significant difference in ?CT between carcinoma tissues and paracancerous tissues(P<0.05),and there was a statistically significant difference in ?CT between carcinoma tissues and marginal tissues(P<0.05).There was no significant difference in ?CT between marginal tissues(P>0.05).This indicates that the expression of SLC5A8 gene in carcinoma tissues is significantly lower than that in paracancerous tissues and marginal tissues.Conclusion The methylation level of SLC5A8 gene in thyroid carcinoma tissue is significantly higher than that in paracancerous and its marginal tissue.The expression level of SLC5A8 gene in thyroid carcinoma tissue is significantly lower than that in paracancerous tissue and marginal tissue.It suggests that the silencing of SLC5A8 gene may be related to its methylation,and the silencing of SLC5A8 gene expression may be involved in the occurrence of thyroid carcinoma.
