Objectives Researching the effects of gene SLC5A8 on tumor occurrence and development was gradually more and moreobvious, and it could be used as an important indicator of tumor early diagnosis and evaluation of prognosis.At present, the research on the gene SLC5A8 was still in the initial stage in China.Through exploring the expression of gene SLC5A8 in colorectal cancer tissue, methylation status and their correlation,it would lay the foundation of the further explore of the gene SLC5A8 on tumor cell apoptosis and study on the correlation of gen SLC5A8 e methylation in the prognosis of colorectal cancer patients.Method The samples gathered from 40 colorectal cancer patients, including cancers,tissue adjacent to carcinoma and normal tissues.(1) Extracted RNA,and gene SLC5A8 m RNA expression level was detected by real-time fluorescent quantitative PCR(2) Extracted DNA, then MSP method was used to detect colorectal cancer tissue, the tissue adjacent to carcinoma and normal tissues of gene SLC5A8 methylation level. SPSS16.0 statistical softwwere was used to processes data, real-time fluorescent quantitative PCR results was analyzed by variance analysis, the other results were analyzed by Fisher's exact probability method or chi-squwere test, inspection standard for P < 0.05 represents the difference was statistically significant.Results(1) Analyzed the differentials(?CT) of cycle numbers of RT-PCR, there were visible differences between the groups.Further analysis between cancer tissue and paracancerous tissueis that there was statistical significance difference in ?CT(t=1.71, P <0.05), and between cancer tissue and normal tissue,?CT differences was statistical significance(t=2.38, P < 0.05), between cancerous tissue and normal tissue, futher, ?CT differences without statistical significance(t=0.67(P > 0.05).Gel electrophores was showed that only 30% of 40 cases of colorectal cancer tissue(12/40) showed obvious stripe;Corresponding in colorectal tissue adjacent to carcinoma, 75%(30/40) showed obvious stripe;Corresponding normal colorectal tissues in 77.5%(35/40) showed obvious stripe.Colorectal cancer tissue, the tissue adjacent to carcinoma and normal tissue positive rate of transcription of gene SLC5A8 m RNA has obvious differences between and the difference was statistically significant(2c= 17.06, P < 0. 05).Further analysis showed that tissue adjacent to carcinoma and normal positive rate of transcription of gene SLC5A8 m RNA was no difference between groups, and both have significant differences with cancerous tissue.(2)and 40 cases of tissue gene SLC5A8 methylation PCR, according to the results of 40 cases of colorectal cancer tissue in 72.5%(29/40) showed obvious stripe;Corresponding in colorectal tissue adjacent to carcinoma, 10%(4/40) showed obvious stripe;Only 5% of the corresponding normal colorectal tissues showed obvious stripe(2/40).Colorectal cancer tissue,tissue adjacent to carcinoma and normal tissues methylation positive rate of gene SLC5A8 had obvious differences, and the difference was statistically significant(2c= 34.68, P < 0.05).Gene SLC5A8 methylation status and clinical features of patients with colorectal cancer,according to the analysis of the relationship between gene SLC5A8 methylation in colorectal cancer patients' age, gender, position, and clinical stage.Conclusion(1)The expression of gene SLC5A8 in the cancer tissue is significantly lower than tissue adjacent to carcinoma and normal tissue; the phenomenon of gene SLC5A8 silencing expression may be involved in the occurrence of colorectal cancer.(2) The methylation of gene SLC5A8 in the cancer tissue is significantly higher than tissue adjacent to carcinoma and normal tissue; the methylation of gene SLC5A8 may had relationship with the occurrence of colorectal cancer.(3)the research of methylation of gene SLC5A8 may provided new targets for the targeted treatment of colorectal cancer.
