Objective To investigate the expression of SLC5A8 gene in colorectal cancer and its methylation phenomenon.To further explore the role of SLC5A8 gene in the development of colorectal cancer,and to investigate the prognosis of colorectal cancer patients with SLC5A8 gene methylation Lay the foundation.Methods In this study,50 patients with colorectal cancer were collected from 2015 to 2016,26 patients with rectal cancer,24 patients with colon cancer,29 males and 21 females,aged 52-80 years,mean age 60.5 years,Collect general information.Collected colorectal cancer tissue,adjacent tissue(0.5cm away from the edge of the tumor),normal tissue(5cm away from the edge of the tumor),stored in liquid nitrogen in reserve.All patients underwent any radiotherapy and chemotherapy and immunotherapy.The extracted DNA was subjected to bisulfite modification according to the Epi Tect Fast DNA Kit kit instructions for methylation-specific PCR using the Epi Tect?MSP kit.After amplification,the amplified product was subjected to electrophoresis.The agarose gel plate was placed in a fully automatic gel imager,developed under ultraviolet light,photographed,and the experimental results were recorded.All the data were analyzed by SPSS19.0 statistical software.Quantitative data were analyzed by ANOVA.Fisher's exact probability method or chi-square test was used to test the data.P <0.05 was considered statistically significant.Results There was no significant difference in the positive rate of SLC5A8 methylation between the age groups(P> 0.05).There was no significant difference between the sex groups(P> 0.05).The methylation of SLC5A8 gene in 50 cases showed that 72%(36/50)of the cancerous tissue showed 10%(5/50)of the adjacent product.Colorectal tissue was 2%(1/50).The methylation of the cancer tissue was significantly more than that in the adjacent tissues and normal colorectal tissues.There was a significant difference in the positive rate of methylation of SLC5A8 gene in colorectal cancer tissues,adjacent tissues and normal tissues,and the difference was statistically significant(x2 =-34.68,P <0.05).The positive rate of methylation of SLC5A8 gene was significantly higher than that of adjacent tissues(72% vs.10%,x2 = 32.24,P <0.05).The normal tissues(72% vs.2%,x2 = 45.88(P <0.05),but there was no significant difference in the positive rate of methylation of SLC5A8 gene between adjacent tissues and normal tissues(10% vs.2%,P> 0.05).Of the 36 cases of SLC5A8 methylated colorectal cancer,2 cases detected unmethylated SLC5A8 gene,which was partial methylation.In SLC5A8 gene,there was no methylated colorectal cancer tissue.3 cases also did not detect unmethylated SLC5A8 gene.3.SLC5A8 gene methylation occurred in 36 of 50 colorectal cancer tissues.34 cases(94.44%)of these 36 positive samples did not detect SLC5A8 expression.Twelve cases(85.71%)were detected in 14 samples of SLC5A8 methylation negative,and there was significant difference(x2 = 26.130,P <0.05).The expression of SLC5A8 was negatively correlated with its methylation status(r =-0.682).conclusion 1.The silencing of SLC5A8 gene expression may be involved in the occurrence of colorectal cancer,and its expression in cancer tissues is significantly lower than that in adjacent tissues and normal tissues.2.The silencing of SLC5A8 gene may be related to its methylation,and the methylation of gene may be significantly higher in cancer tissues than in adjacent tissues and normal tissues.3.The SLC5A8 gene plays an important role in the development and progression of colorectal cancer.The methylation of SLC5A8 gene has laid a foundation for the study of prognosis of patients with colorectal cancer.
