Objective:Through previous studies,We found that the expression of CADM1 decreased or was absent in bladder urothelial carcinoma,and was negatively correlated with pathological grade.When the expression of CADM1 was up-regulated the proliferation and invasiveness of tumor cells were significantly reduced.While the expression of CADM1 was down-regulated,the proliferation and invasion of tumor cells increased significantly.CADM1 could inhibit the transformation of G1/S phase of tumor cell cycle through Rb/E2 F signaling pathway,and then inhibited the formation of tumor in hepatoma cells.The purpose of this study is to compare the expression of CADM1,RB and E2F1 in bladder urothelial carcinoma and normal bladder mucosa by collecting clinical data of the patients with bladder urothelial carcinoma.In this study,we want to verify the correlation among the three factors and the correlation between these factors and the clinicopathological data,which lays a theoretical foundation for the further study of the role of this signaling pathway in the development of bladder urothelial carcinoma.Methods:The specimens of 86 cases of bladder urothelial carcinoma and 24 cases of normal bladder mucosa were collected.QPCR method was used to detect the expression of mRNA of CADM1,Rb,E2F1 in cancer tissues and normal tissues,and the expression of three proteins was detected by westernblot method.The relationship among the three proteins and the relationship between the expression of these proteins and the clinicopathological data were analyzed.Results:1.The results of qPCR show that the average expression of CADM1 mRNA in bladder urothelial carcinoma tissues is 0.754±0.225,which is lower than normal bladder mucosa(1.340±0.180,P=0.024).The average expression of Rb mRNA in bladder urothelial carcinoma tissues is 0.725±0.247,which is lower than normal bladder mucosa(1.476±0.248,P=0.033),while the average expression of E2F1 mRNA in bladder urothelial carcinoma tissues is 1.443±0.304,which is higher than normalbladder mucosa(0.463±0.178,P=0.041),all of the differences is statistically significant.2.The results of WB show that the average expression of CADM1 in bladder urothelial carcinoma tissues is 0.342±0.105,which is lower than normal bladder mucosa(0.538±0.185,P=0.008).The average expression of Rb in bladder urothelial carcinoma tissues is 0.369±0.154,which is lower than normal bladder mucosa(1.476±0.248,P=0.033),while the average expression of E2F1 mRNA in bladder urothelial carcinoma tissues is 1.443±0.304,which is higher than normal bladder mucosa(0.477±0.188,P=0.015),all of the differences is statistically significant.3.In the 86 bladder urothelial carcinoma specimens,Spearman correlation analysis shows that there is a positive correlation between the expression of CADM1 and Rb(r=0.785,P=0.017),while the correlation between the expression of CADM1 and E2F1(r=-0.534,P=0.008)and the correlation between the expression of Rb and E2F1(r=-0.457,P=0.003)are negative.4.In the correlation analysis between the expression of these three proteins and clinicopathological features,we do not find significant correlation between the expression of them with sex,age,tumor size and tumor number of the patients.The expression of CADM1 in low grade bladder urothelial carcinoma tissues is0.376±0.009,which is higher than high grade(0.327±0.005,P<0.01).The expression of Rb in low grade bladder urothelial carcinoma tissues is 0.394±0.006,which is higher than high grade(0.359±0.003,P<0.01),while the expression of E2F1 in low grade bladder urothelial carcinoma tissues is 0.553±0.017,which is lower than high grade(0.609±0.012,P=0.012).Conclusions:1.In bladder urothelial carcinoma tissues,the expression of CADM1 and Rb is significantly decreased,and the expression of E2F1 is significantly increased.2.In bladder urothelial carcinoma,there is a positive correlation between the expression of CADM1 and Rb.The expression of CADM1 and Rb have a negative correlation between the expression of E2F1,meanwhile the correlation between the pathological grade and the expression of CADM1 and Rb is negative.The expression of E2F1 has a positive correlation with pathological grade.3.Through our study,we find the expression changes and clinicopathological characteristics of CADM1,Rb and E2F1 in bladder urothelial carcinoma,which will lay a theoretical foundation for our further study of the role of CADM1 and Rb/E2F1 signal pathway in the development of bladder urothelial carcinoma.
