Objective:To investigate the expression of Skp2 and Spl in different clinical stages and pathological grades of bladder urothelial carcinoma and their association in the same bladder cancer tissues,and the relationship between the recurrence of bladder cancer.Method:A total of 69 patients were selected from January 2012 to December 2015 in Yanbian University Hospital,who were diagnosed with bladder urothelial carcinoma after surgery.In addition,20 cases of non-tumor bladder mucosa were used for control study.The patients were not given chemotherapy,radiotherapy and immunotherapy before surgery.The expression of Skp2 and Sp1 protein in 69 cases of bladder cancer and 20 cases of non-tumor bladder mucosa were detected by immunohistochemistry PV6000 method.To analyze the expression of Skp2 and Spl in different clinical stages and pathological grades of bladder urothelial carcinoma and its clinical significance.Result:(1)The positive expression rates of Skp2 in 69 cases of bladder cancer and 20 cases of non-tumor bladder mucosa were 69.6%and 10.0%,with significant differences(P<0.05).The positive expression rates of Skp2 in superficial bladder cancer group and invasive bladder cancer group were 57.1%and 88.9%,with significant differences(P<0.05).The positive expression rates of Skp2 in low-grade bladder cancer group and high-grade bladder cancer group were 50.0%and 80.0%,with significant differences(P<0.05).The positive rates of Skp2 in patients with recurrent and non-relapsed patients were 85.3%and 54.3%,with significant differences(P<0.05).(2)The positive expression rates of Spl in 69 cases of bladder cancer and 20 cases of non-tumor bladder mucosa were 81.2%and 15.0%,with significant differences(P<0.05).The positive expression rates of Skp2 in superficial bladder cancer group and invasive bladder cancer group were 71.4%and 96.3%,with significant differences(P<0.05).The positive expression rates of Skp2 in low-grade bladder cancer group and high-grade bladder cancer group were 58.3%and 93.3%,with significant differences(P<0.05).The positive rates of Skp2 in patients with recurrent and non-relapsed patients were 91.2%and 71.4%,with significant differences(P<0.05).(3)There was a positive correlation between the expression of Skp2 and Sp1 in the same bladder cancer tissues(P<0.05).Conclusion:(1)Skp2 and Spl are closely related to the clinical stage and pathological grade of bladder cancer,suggesting that Skp2 and Spl may be involved in the process of bladder cancer cell proliferation,invasion,metastasis and recurrence.The expression of Skp2 was positively correlated with the expression of Spl in bladder cancer,suggesting that Sp1 may promote the occurrence and development of bladder cancer by activating Skp2.Combined detection of Skp2 and Spl has a certain value in assessing the prognosis of bladder cancer.
