Objective To observe the effect of exercise training combined with fluoxetine on the degree of depression and the expression of brain-derived neurotrophic factor(BDNF)and tropomyosin receptor kinase B(Trk B)protein in hippocampus of post-stroke depression rats,and to explore the possible mechanism of combined treatment for post-stroke depression.Methods Forty eight male Wistar rats were randomly divided into the sham operation group,the model group,the fluoxetine group and the combined treatment group(n=10 for each group).All except the sham operation group were given the middle cerebral artery occlusion(MCAO)using the intraluminal tread method and a chronic unpredictable stress(CUMS)was applied.The model group and the sham operation group were not giving any training.Fluoxetine group was given fluoxetine treatment,combined treatment group received exercise training and fluoxetine treatment,and these treatments lasted for 28 days.After 14 days and 28 days treatment,four groups of rats were performed behavioral experiments(sucrose consumption test?forced swimming test and body weight test).The expression of BDNF and Trk B in the left hippocampus were detected by Western blot.Results There were significant differences between the model group and the sham operation group.After 14 days and 28 days of treatment,behavioral tests showed that the degree of depression in fluoxetine group and combined treatment group were significantly lower than that in the model group.At same time point,the expression of BDNF and Trk B in the left hippocampus of the combined treatment group was significantly higher than that of the fluoxetine group.Conclusion Exercise training combined with fluoxetine can improve the depressive state of rats with post stroke depression.
