Objective For optimizing the extraction process of the active ingredients of Panax notoginseng,screen out the best preparation technology of Panax notoginseng dropping pills,establish the determination method of Panax notoginseng dropping pills,and study the electrochemical fingerprint of Panax notoginseng and its extracts,fingerprinting on the quality of Panax notoginseng extract from the preparation of the whole process of dropping pills for full control to ensure that the quality of Panax notoginseng pills meet the standards.Methods The orthogonal experiment was used to design with ethanol proportion,solid-liquid ratio and extraction reflux time as the main factors to determine the best extraction technology of Panax notoginseng.The single-factor test method was used to determine the basic conditions of Electrochemical Fingerprinting of Panax notoginseng extracts with the concentration of each solvent,temperture,stirring time,and stirring speed.Using this method,the active ingredients of Panax notoginseng and the amount of Panax notoginseng dripping pills were determined.The Panax notoginseng extract was prepared as a drop pill by solid dispersion technique.The effects of different types of matrix and the ratio of matrix to drug were investigated respectively.Influence,optimize the preparation of the best dropping pills;Determination of ginsenoside Rg1 using high performance liquid chromatography,and to establish a method for the determination of content;using non-linear electrochemical fingerprint technology,induction time for the evaluation of technical indicators,extracts active ingredients and dropping pills were measured.Results Through orthogonal test,the optimal extraction technology of the active ingredients of Panax notoginseng was determined as follows:90%ethanol was used;reflux extraction time was 60 minutes;the solid-liquid ratio was 12 times;the electrochemical fingerprint of Panax notoginseng extract was controlled by temperature at(37±0.02℃),and the contents of H2SO4(1.0 mol/L),MnSO4(0.02 mol/L)and the acetone(0.6 mol/L)were kept in the reaction vessel at for 15 minutes.Electromagnetic stirrer speed of 500 r/min,and then 10ml of KBrO3(0.2 mol/L)solution was added to record the electrochemical fingerprints.The optimal preparation process of Panax notoginseng dropping pills is that the ratio of drug to matrix is 1:3,the dropping temperature was 90℃,the dropping distance was 6 cm.At the same time,the dropping pills content was determined by HPLC.The linear range of ginsenoside Rg1 was 0.02280.57 mg/ml,r=0.9997.The average recovery was 100.02%with an RDS value of 0.97%.The method is simple,reliable and accurate and can be used for the quality control of Panax notoginseng pills.At the same time,using electrochemical fingerprint determination of content of notoginseng dropping pills,and compared with the classic high-performance liquid chromatography(HPLC),the determination results showed the same trend,this method can assist the content of Panax notoginseng dropping pills.Conclusions The best extraction process of Panax notoginseng was optimized by Orthogonal test,and Panax notoginseng dropping pills were prepared for ease of adminstration.A HPLC method for the determination of dropping pills was established.Meanwhile,electrochemistry fingerprinting method was established to quickly identify and control the quality of Panax notoginseng pills.
