Background: The early diagnosis rate of gastric cancer is low.Further research on the mechanism of gastric cancer occurrence and development,and finding biomarkers for invasion and metastasis of gastric cancer are of great significance for the prevention and treatment for patients with gastric cancer.Objective: To screen the differential expression of long non coding RNA(LncRNA)related to gastric cancer,and to explore the new target for the diagnosis and treatment of gastric cancer.Method: The microarray data associated with gastric cancer was downloaded in Gene Expression Omnibus(GEO),the differential expression of long nocoding RNA was analyzed with R software,and gene annotation and clustering were performed in DAVID database.GES-1,BGC-823 cells were cultured with 5ng/ml TGF-? with the aim to stimulate the cells and evaluate the changes of cell migration proliferation and invasiveness after establishing EMT cell model.The relationship between epithelial mesenchymal marker and expression level of Lnc RNA MT1 JP was analyzed by using Sprearman's test.Results:The microarray GSE103236 involving 10 normal tissues and 9 gastric cancer tissues was analyzed.Based on the data from microchip,65 differentially expressed genes including40 up-regulated Lnc RNAs and 25 down-regulated Lnc RNAs were screened.The results of gene enrichment analysis showed that most of the differentially expressed genes were enriched in cell matrix and adhesion molecules.Cell proliferation,migration and invasion ability of GES-1,BGC-823 induced by TGF-? were significantly enhanced(p<0.01).The expression level of Lnc RNA MT1 JP and E-cadherin were significantly decreased in BGC-823 cell line.Significantly,the expression level of N-cadherin and Vimentin were increased in TGF-? induced EMT group(p<0.01).Conclusions: The expression of gastric cancer related LncRNA is in different levels.Lnc RNA MT1 JP may be involved in the process of gastric cancer invasion and metastasis.The low expression level of Lnc RNA MT1 JP can promote the EMT process of gastric cancer cells.It can be an important target for invasion and metastasis of gastric cancer.Further exploration of its related mechanisms is expected,which would become a new target for the diagnosis and treatment for gastric cancer.
