Background and ObjectiveEsophageal squamous cell carcinoma(ESCC)is now the world's leading cause of cancer death of sixth,which most of the patients with esophageal cancer belongs to developing countries.The rate of incidence and death of esophageal cancer in China has reached 50% of the world.Esophageal cancer has gradually become one of the main burden of residents in remote and backward areas in the midwest of China due to deviation diet and economic reasons.The current research shows that the 5 year survival rate of esophageal cancer is only 15%~25%.For early esophageal cancer patients,however,the 5 year survival rate can reach 40% after effective individualized treatment.Therefore,studying the etiology of esophageal cancer,especially the mechanism of early esophageal cancer is much critical.S100A8,which belongs to one of the S100 family,is closely related to the occurrence and development of tumor,but its specific mechanism of inducing tumor cell apoptosis remains unclear.The study consists of two parts.Firstly by using real-time PCR and immunohistochemical technique to study the expression of S100A8 in esophageal squamous cell carcinoma and study the relationship between clinical datas of patients with ESCC and expression of S100A8.Secondly the ability of migration and invasion of EC9706 after being treated with S100A8 were detected by using MTT,cell migration assay and tumor invasion assay,thus further study on the relationship between S100A8 and metastasis and invasion of ESCC in order to provide a new direction to the process of the transfer and invasion of ESCC.Part ?: Expression of S100A8 in esophageal squamous cell carcinoma tissuesMethods 1.By using real-time PCR and immunohistochemical technique,the expression of S100A8 in normal esophageal tissues,non-cancerous tissues and esophageal squamous cell carcinoma tissues of the esophagus was preliminarily studied.2.Analyzing the relationship between clinical datas of patients with ESCC and expression of S100A8.Results 1.Real-time PCR and immunohistochemistry showed that the expression of S100A8 m RNA and S100A8 protein in normal esophageal tissues,non-cancerous tissues and esophageal squamous cell carcinoma tissues of the esophagus gradually decreased,and the difference were statistically significant(P<0.05).2.The expression of S100A8 in esophageal squamous cell carcinoma patients was not significantly different from that of in terms of sex,tumor differentiation and tumor volume,but it was related to the age,TNM staging and lymph node metastasis of esophageal squamous cell carcinoma,which the difference was statistically significant(P<0.05).Part?: The impact of S100A8 on the ability of invasion and metastasis of esophageal squamous cell carcinomaMethods 1.The proliferation capacity of S100A8 to EC9706 cells in esophageal squamous cell carcinoma was detected by MTT assay.2.Detection of migration capacity of EC9706 cells in squamous cell carcinoma of the esophagus after S100A8 treatment by cell migration assay and tumor invasion assay.3.Detection of invasiveness migration capacity of EC9706 cells in squamous cell carcinoma of the esophagus after S100A8 treatment by tumor invasion assay.Results 1.S100A8 can inhibit the proliferation capacity of esophageal squamous cell carcinoma EC9706 cells,while at the time of 24 h,concentration of 20?g/ml inhibit the most.2.S100A8 can inhibit the migration of EC9706 cells in the squamous cell carcinoma of the esophagus,and the capacity of inhibition increases with the increase of concentration.3.S100A8 can weaken the invasiveness of EC9706 cells in the squamous cell carcinoma of the esophagus.Conclusion1.The expression of S100A8 m RNA and S100A8 protein in normal esophageal tissues,non-cancerous tissues and esophageal squamous cell carcinoma tissues of the esophagus gradually decreased,which shows that S100A8 is closely related to the development and invasion of esophageal squamous cell carcinoma.2.The expression of S100A8 in esophageal squamous cell carcinoma patients was not significantly different from that of in terms of sex,tumor differentiation and tumor volume,but it was closely related to the age,TNM staging and lymph node metastasis of esophageal squamous cell carcinoma,which the difference was statistically significant,which further suggested that S100A8 may be one of the important markers in the development of squamous cell carcinoma of the esophagus.3.Through MTT assay,cell migration assay and tumor invasion assay,S100A8 shows the inhibitory effect on proliferation and invasion and metastasis of EC9706 of esophageal squamous cell carcinoma,which provides a new direction for early prevention and treatment of esophageal squamous cell carcinoma.
