| Backgrounds:Esophageal squamous cell carcinoma(ESCC) is the most common and aggressive malignant tumour in digestive tract, The incidence tends to increase with the patients'ages dropping in recent years. China has one of the highest-incidences and death rate of esophageal cancer in the world, predominantly esophageal squamous cell carcinoma. Patients with ESCC generally have a worse prognosis than those with other digestive trcat cancers even after curative surgical resection,primarily because of the frequency of extensive local invasion and regional lymphnode metastasis at presentation.The latest research findings suggests that the extracellular matric (ECM) plays an important role in the invasion and metastasis.MMP-9 can dissolve collage IV, which is the main ingredient of extracellular matrix and basement membrane, and make the tumor cells break their original sites to bring about the invasion and metastasis. Recent findings indicate CD-147,which belongs to immunogloblulin superfamily, can induce the release of MMPs in multiple ways and then help the invasion and metastasis of tumor cells. The capacity of invasion and metastasis is closely related to the capacity of CD-147 to induce MMPs.Purposes:To investigate the expressions of protein and messenger RNA of matrix metalloproteinase-9(MMP-9) and CD-147 in human esophageal squamous cell carcinoma to explored the function and interaction of MMP-9 and CD-147 in the invasion and metastasis of esophageal squamous cell carcinoma. By this study we hope to find reasonable and effective diagnose and treating scheme, therefore, increase the survival ratio of the patients.Methods:1. we evaluated MMP-9,CD-147 proteins expression and relationship to clinicopathologic characteristics in esophageal squamous cell carcinoma and esophageal epithelial cells from normal tissues adjacent to the tumours from 57 patients using immunohistochemistry S-P methods.2. we evaluated MMP-9,CD-147 messenger RNA expression and relationship to clinicopathologic characteristics in esophageal squamous cell carcinoma and esophageal epithelial cells from normal tissues adjacent to the tumours from 57 patients using Reverse transcription-polymerase chain reaction(RT-PCR) methods.Results:1. The expression of MMP-9 protein in esophageal squamous cell carcinoma and esophageal epithelial cells from normal tissues adjacent to the tumours.The positive rate of MMP-9 protein was 82.456% in ESCC and 29.825% in the compared tissues, with significant difference between them(P<0.01). The expression of MMP-9 protein was correlated with the depth of tumor infiltration and lymph node metastasis, but not correlated with degree of differentiation, patient age ,patient sex ,tumour size. The positive rate of MMP-9 protein in mucous and submucosal interstitial cells around cancerous lesions was 33.333%,but in the muscular layer and adventitia was 85.185%, with significant difference between them(P<0.05). The positive rate of MMP-9 protein in lymph node metastasis was 93.333%,but it was 78.571 % without lymph node metastasis, there was significant difference between them(P<0.05).2. The expression of CD-147 protein in esophageal squamous cell carcinoma and esophageal epithelial cells from normal tissues adjacent to the tumours.The positive rate of CD-147 protein was 64.912% in ESCC and 8.771% in the compared tissues, with significant difference between them(P<0.01). The expression of CD-147 protein was correlated with the depth of tumor infiltration and lymph node metastasis, but not correlated with degree of differentiation, patient age ,patient sex ,tumour size. The positive rate of CD-147 protein in mucous and submucosal interstitial cells around cancerous lesions was 0,but in the muscular layer and adventitia was 68.519%, with significant difference between them(P<0.05). The positive rate of CD-147 protein in lymph node metastasis was 93.333%,but it was54.762% without lymph node metastasis, there was significant difference between them(P<0.05).3. The expression of MMP-9 mRNA in esophageal squamous cell carcinoma and esophageal epithelial cells from normal tissues adjacent to the tumours.The positive rate of MMP-9 mRNA was 71.923% in ESCC and 26.316% in the compared tissues, with significant difference between them(P<0.01). The expression of MMP-9 mRNA was correlated with the depth of tumor infiltration and lymph node metastasis, but not correlated with degree of differentiation, patient age ,patient sex ,tumour size. The positive rate of MMP-9 mRNA in mucous and submucosal interstitial cells around cancerous lesions was 0,but in the muscular layer and adventitia was 75.926%, with significant difference between them(P<0.05). The positive rate of MMP-9 mRNA in lymph node metastasis was 93.333%,but it was 64.286 % without lymph node metastasis, there was significant difference between them(P<0.05).4. The expression of CD-147 mRNA in esophageal squamous cell carcinoma and esophageal epithelial cells from normal tissues adjacent to the tumours.The positive rate of CD-147 mRNA was 68.421% in ESCC and 21.053% in the compared tissues, with significant difference between them(P<0.01). The expression of CD-147 mRNA was correlated with the depth of tumor infiltration and lymph node metastasis, but not correlated with degree of differentiation, patient age ,patient sex ,tumour size. The positive rate of CD-147 mRNA in mucous and submucosal interstitial cells around cancerous lesions was 0,but in the muscular layer and adventitia was 72.222%, with significant difference between them(P<0.05). The positive rate of CD-147 mRNA in lymph node metastasis was 86.667%,but it was61.905% without lymph node metastasis, there was significant difference between them(P<0.05).5. the correlateion of MMP-9 and CD-147The simultaneous positive rate of the CD-147 protein and the MMP-9 protein was 63.158%(36/57) ,but the simultaneous negative rate was 15.789%(9/57),there was a positive significant correlateion between the expression of CD-147 and MMP-9 protein in ESCC.The X~2=17.907,and the spearman coefficient was0.451 ,P<0.001. The simultaneous positive rate of the CD-147 mRNA and the MMP-9 mRNA was 68.421%(39/57) ,but the simultaneous negative rate was 10.526%(6/57),there was a positive significant correlateion between the expression of CD-147 and MMP-9 mRNA in ESCC.The X~2=4.534,and the spearman coefficient was0.282,P<0.05. Conclusions:1. MMP-9 and CD-147 did highly expressed on esophageal squamous cell carcinoma but not not on normal esophageal tissue.2. The expression of MMP-9 and CD-147 was correlated with the depth of tumor infiltration and lymph node metastasis, but not correlated with degree of differentiation, patient age ,patient sex ,tumour size.the expression of MMP-9 and CD-147 is close relationship with the invasion and metastasis of ESCC.3. there was a positive correlateion between the MMP-9 and CD-147 in ESCC.The expression of CD-147 may be an inducer of MMP-9,which could enhance the expression of MMP-9. |
