Objective: To study the changes of taurine content in liver of children with biliary atresia(BA)and BA mouse model.Method: Twenty-four cases of liver specimens from children with BA were treated as experimental group and eight cases of liver specimens from children with benign tumor as control group.Sixteen newborn SPF mice within 12 hours were divided into two groups randomly.The mice of experimental group(n=8)received RRV by intraperitoneal(ip)and the mice of control group(n=8)received the same dose of saline by ip.We detected the histopathological features of liver by HE staining and the taurine content in liver by ELISA kit.Results: HE staining showed that there were different levels of inflammatory cell infiltration in the portal area of the BA model and a small amount of dot necrosis and less intrahepatic bile duct hyperplasia.ELISA showed that the content of taurine in the liver of BA was significantly higher than that in the control group(P <0.05).Similarly,the content of taurine in the liver tissue of the BA mouse model was also significantly increased and the difference between this group was statistically significant(P <0.05).Moreover,the content of taurine in the liver of mice with different time course of BA was found to significantly increased and followed by a sharply decreasing.The difference was statistically significant(P <0.05).Conclusion: The content of taurine in liver tissue of children with BA was significantly increased;The content of hepatic taurine in experiment mice was also significantly increased but it followed by a sharply decreasing.Objective: To study the expression of Tau T in liver tissue and to explore the mechanism of influencing the content of taurine in BA liver tissue.Method: Twenty-four cases of liver specimens from children with BA were treated as experimental group and eight cases of liver benign tumor liver as control group.The liver samples of BA model in 3,7,14,21 days were taken as experimental group,At the same time,the hepatic specimens of the mice with the same age were taken as the control group.The Tau T protein express in liver was detected by immunohistochemical staining.The expression of Tau T protein was detected by Western Blot.The transcription of Tau T m RNA was detected by RT-PCR.Results: In the liver of children with BA,The Tau T gene express in liver was significantly increased,detected by Western Blot and RT-PCR,the difference was statistically significant(P <0.05).In the model of BA mice,the expression of Tau T protein and m RNA in the liver was also significantly higher than that in the control group(P <0.05).The results of Western Blot and RT-PCR showed that the difference was significant.The expression of Tau T protein and m RNA in the liver tissue of the BA mice increased firstly and then decreased,the difference was statistically significant(P <0.05).Conclusion: The expression of Tau T in the liver tissue of children with BA and the mouse model was increased,and the expression of Tau T in the liver tissue of mice with different degrees of BA increased first and then decreased.Objective: To study the expression of CSAD in liver tissue and to explore the mechanism of influencing the content of taurine in BA liver tissue.Method: Twenty-four cases of liver specimens from children with BA were treated as experimental group and eight cases of liver benign tumor liver as control group.The liver samples of BA model in 3,7,14,21 days were taken as experimental group,At the same time,the hepatic specimens of the mice with the same age were taken as the control group.The expression of CSAD protein was detected by Western Blot,and the expression of CSAD m RNA in liver was detected by RT-PCR.Results: In the liver of children with BA,the expression of CSAD was slightly increased by Western Blot and RT-PCR,but compared with the control group,The difference was not statistically significant(P> 0.05).in the liver of mice with BA,the results of Western Blot and RT-PCR showed that CSAD protein and m RNA were slightly higher than those in control group,The difference was not statistically significant(P> 0.05).There was no significant difference in the expression of CSAD in liver tissue of BA mice with different agesConclusion: The expression of CSAD does not significantly increased In the liver of children with BA patients and BA mouse models.The expression of CSAD does not significantly increased In the liver of mice with BA.
