Objective: To confirm that flavored sijunzi decoction can delay the aging of mice and to study the mechanism of anti-aging effect of flavored sijunzi decoction by regulating mTOR signaling pathways.Porpose: A total of 30 SPF male kunming mice at the age of 8 weeks,which were fed with adaptive feeding for 3 days,were randomly divided into the low dose group,high dose group and the control group,with each group of ten only.The low dose group every day was given flavored sijunzi decoction(0.112g/d)to fill the stomach,and was intraperitoneal injection of D-gal solution(0.2g/Kg.D);The high dose group every day was given flavored sijunzi decoction(0.28g/d)to fill the stomach,and was intraperitoneal injection of D-gal solution(0.2g/Kg.D);The control group every day was given normal saline lavage volume to fill the stomach,and was intraperitoneal injection of D-gal solution(0.2g/Kg.D).The all mice were fed to 42 days and then sacrificed.The brain,kidney,spleen organs of three groups of mice were taken for HE staining,and the morphological changes of the organs of three groups were observed.At the same time,the molecular biology method was used to detect PI3K(mTOR signaling pathways of upstream signal factor),p70S6K(mTOR signaling pathways downstream of effect factors)and 4E-BP1(mTOR signaling pathways downstream of effect factors)three indicators,and the date were analyzed statistically.The results showed that P<0.05 was statistically significant.The resulis: 1)Histomorphology result: Low dose group,high dose group compared with control group respectively,the changes of hismorphology of brain,kidney,spleen tissues of mice were lighter,and the degree of change was different.In the control group,the hippocampus of the brain tissue of mice showed different degrees of neuronal swelling,loss of neurons,nuclear pycnosis;The kidney tissue can see atrophy of glomerular,the renal tubular cell swelling or necrosis,and the renal interstitial fibrosis lesions;There was no clear demarcation of the white pulp and red pulp in the spleen tissue,the spleen was significantly smaller in Liang Zheng-Sheng and the fibers in the red pulp were thicker.2)Molecular biology results: low dose group,high dose group compared with control group respectively,the expression of PI3 K and p70S6 K in brain,kidney and spleen tissues of mice decreased(P<0.05);Compared to low dose group and high dose group,there was no difference in the degree of the expression of PI3 K and p70S6 K in brain,kidney and spleen tissues of mice(P >0.05).Compared with the three groups,the degree of the expression of 4E-BP1 in brain,kidney and spleen tissues of mice had no difference(P>0.05).Conclusion: 1)Flavored sijunzi decoction can delay the aging of mice.2)The mechanism of anti-aging effect of Flavored sijunzi decoction may be related to inhibition of mTOR signaling pathways,decreasing the expression of PI3 K and p70S6 K level.
