Objectives Through the P.g LPS stimulation and bacterium orthodontic wire ligature method to establish experimental periodontitis model in SD rats,modeling and on the basis of treatment success after removal of ligation silk,take the rat peripheral blood(PB)samples,gum tissue and jaw.In gingival tissue and peripheral blood were observed based on CCL2,c-Jun,IL-1? before and after treatment of expression,The relationship between the changes of their content and periodontal tissue inflammation was analyzed,and the effects of them on periodontitis were further explored.In order to explore whether CCL2,c-Jun and IL-1? can be used as indicators to reflect periodontal inflammation and whether changes in inflammatory factors may be related to the occurrence of systemic diseases.Methods Selected 6 weeks of age 15 male SD rats were randomly divided into three groups:blank control group(CN),experimental periodontitis group(EP),experimental periodontitis + local debridement treatment group(SRP),each group 5 only.The rat model of periodontitis was established by orthodontic ligation wire ligation + P.g Lps injection.There was no intervention in group CN,and the SRP group was treated with ligation removal and local debridement once a day for 2 weeks.Rats in groups CN,EP and SRP were killed at 0,2 and 4 weeks after the completion of the modeling,and gingival tissue,jaw bone samples and peripheral blood were collected from each group.The BI of the experimental teeth of each group was examined by a trained person.Continuous slices of periodontal tissue were made in the maxilla samples.Through HE staining,TRAP staining,IHC,ELISA and PCR,the expression of CCL2,c-Jun and IL-1? in periodontal tissues,gingival tissues and PB serum was observed.Resulte 1.In the experimental periodontitis group,the gingival tissue of rats presented dark red color,accompanied by edema and congestion.The gingival redness and swelling of rats in group SRP were improved,compared with the group CN(P < 0.05).No obvious inflammatory changes were observed in the maxillary first molar of rats in group CN.2.Observed the histologic section,The group EP has obvious inflammatory change,connective tissue layer is a large number of inflammatory cells infiltration,collagen fibers degeneration,edema,alveolar bone absorption area is a large number of boneresorption lacunae,including visible active OCs.In group SRP,the inflammation of periodontal tissue was reduced,the OCs were decreased,and the absorption of alveolar bone was still,indicating the new fibrous connective tissue.Large collagen fibers were seen in group CN.3.IHC staining showed that the expression levels of CCL2,c-Jun,and IL-1? in group EP gingival tissues were significantly higher than those in CN group(P<0.05),while the expression levels of CCL2,c-Jun,and IL-1? in group SRP were significantly lower than group EN(P<0.05).4.The ELISA test results showed that CCL2 and IL-1? were significantly higher in the blood serum of the periodontitis,and the group SRP was significantly lower than the EP group,but the difference was not statistically significant(P>0.05).5.The results of PCR showed that the expression of CCL2,c-Jun and IL-1? mRNA in gingival tissue and peripheral white blood cells of group EN was significantly higher than that in group CN(P<0.05),while the group SRP was significantly lower than group EN(P<0.05).conclusion 1.ligation of +P.g LPS injection can successfully establish experimental periodontitis model in a short period.This model is an effective means to study the pathological changes of periodontitis.It provides a scientific basis for the clinical study of periodontitis,and it is of great significance for the treatment of periodontitis.2.CCL2,c-Jun and IL-1? levels are closely related to the degree of periodontal inflammation,indicating that the level of CCL2,c-Jun and IL-1? in gingival tissue and peripheral blood can be used as an indicator of periodontal tissue inflammation.3.During the development of periodontitis,CCL2,c-Jun and IL-1? not only increase in gingival tissue,but also change in the whole body circulation,indicating that inflammatory factors can be related to the occurrence of systemic diseases.
