?-lactam of many natural and synthetic ?-lactam antibiotics are the core of the structure,such as penicillin and cephalosporin,penicillium carbon alkene and monocyclic ?-lactam,and as a kind of very useful in organic synthesis intermediates.Asymmetric Kinugasa reaction used for synthesis of different structural optical pure ?-lactam has many attractive advantages,such as raw materials easily accessible,high functional group tolerance and comply with the atom economy.Therefore,asymmetric Kinugasa reaction has attracted many scientists to conduct research and have achieved good results.A lot of these studies,however,tend to generate cis ?-lactam,recently,the demand for trans ?-lactam gradually increase,because they have the potential to be used as a serine protease inhibitor and a ?-lactamase inhibitor.Therefore,the use of asymmetric Kinugasa reaction for synthesis of high reactivity,excellent diastereoselectivity and good enantioselectivity trans ?-lactam product has great research value.In order to solve this problem,we developed a novel chiral schiff base sheet metal complex to catalytic asymmetric Kinugasa reaction.After a series of optimized reaction condition,we synthesized a series of trans ?-lactam products under the optimal reaction conditions.Results show that the catalytic systems for aromatic alkyne,aromatic nitro compounds and aromatic aldehyde has good performance of chiral induction,trans ?-lactam product yield up to 96%,more than 99:1 to the diastereoselectivity and 95% ee.Numerous studies have reported that the beta lactam derivatives have antibacterial activity outside of a variety of biological activity.However,for antitumor effect especially less resistance to breast cancer research.In this paper,a highly ability of invasion and metastasis of breast cancer cells as the research object.First of all,using crystal violet and Annexin V/PI method to detect the trans ?-lactam product effect on breast cancer cell proliferation and apoptosis.Secondly,using the scratches and Transwell experimental detection trans ?-lactam product effect on breast cancer cell migration and invasion.
