Objective:To observe the differential expression of RBM5 in the brain glioma tissue,and to explore the correlation between the expression level and the malignant degree of glioma.Methods:1 41 cases of human glioma specimens were collected in the Department of neurosurgery in the second hospital of Hebei Medical University for2016-2018 years.4 brain tissue specimens with cerebral hemorrhage or brain injury requiring decompression or fistula were made as the group.The specimens needed for the experiment were divided into two parts.The quality was about 200 mg.Some specimens were fixed in 10% formalin solution.The other part was first placed in 2ml refrigerated tube,and then placed in liquid nitrogen for 3-5 seconds,and then transplanted to-80 degree refrigerator.2 The expression of RBM5 in the experimental group and the control brain tissue was detected by immunohistochemical method,and the difference in the expression between the control group and the experimental group was analyzed.3 The Western blot method was used to detect the expression of RBM5 protein in the experimental group and the control brain tissue,and The relationship between the expression of RBM5 and the pathological grade was analyzed and compared.4 The qRT-PCR method was performed to determine the relative expression of RBM5 mRNA in the brain tissue of the experimental group and the control group,and the expression difference was analyzed and compared.Results:1.Immunohistochemical results: the positive expression of RBM5 was found in 4 control brain tissues,and the positive expression of RBM5 in brainglioma was73.17 %.RBM5 protein is expressed in different levels of glioma cells.The relative expression of RBM5 protein was statistically significant in the control group,the low grade glioma group and the high grade glioma group.2.Western blot results:The relative expression of RBM5 protein was statistically significant in the control group,the low grade glioma group and the high grade glioma group..3.q RT-PCR results: the relative expression of RBM5 mRNA was statistically significant in the control group,the low grade glioma group and the high grade glioma group.Conclusions:1.RBM5 was expressed in the brain glioma tissue,and its expression was decreased with the increase of pathological grade of glioma.2.The expression of RBM5 between low grade glioma and high grade glioma is statistically significant,and is associated with the malignancy of glioma.3..RBM5 may play an important role in the occurrence,development and infiltration of glioblastoma.It may be an important indicator to judge the malignant biological behavior and prognosis of glioma,and may be a new target gene for the treatment of glioma.
