Objective: To investigate the biological effect of calcium-integrin binding protein (CIB) overexpression in human malignant glioma cell line.Methods: (1) CIB gene was transfected into human SHG44 glioma cell line by lipofectamine, and positive clone (SHG44-CIB) was screened by G418. (2) The expression of CIB gene was detected by reverse transcription-polymerase chain reaction (RT-PCR). (3) MTT assay and soft agar colony formation was used to measure the effect of CIB overexpression on growth feature of the transfected glioma cells.Results: CIB gene was successfully transfected into SHG44 cells, and the stable expression cell line was obtained by G418 screening. The expression of CIB mRNA increased in SHG44 cells which was stably transfected with pcDNA3-CIB vector. The overexpression of CIB gene was examined by RT-PCR analysis. Seventy-two hours after transfection, the growth of glioma cells transfected with pcDNA3-CIB vector was significantly inhibited compared with the control cells (P<0.01) by MTT assay. Using soft agar colony formation, we found that cell proliferation rate of SHG44 cells transfected with CIB cDNA was reduced, and showed statistically significant difference(P<0.05).Conclusions: (1) The stable expression cell line(SHG44-CIB) was obtained by G418 selection. The results provide experimental model for further study on the function of CIB in tumor tissues, and for the study of glioma gene therapy. (2) The expression of CIB mRNA increased in SHG44 cells which was stably transfected. By MTT assay and soft agar colony formation, we found that overexpression of CIB gene could suppress the growth of SHG44 malignant glioma cells.
