Objective:In this research,the depressive rats were induced by reserpine,the Simplified Chaihu-Shugan Powder(SCSP)was used as the treatment,the fluoxetine was used as a positive control drug,to observing the depressive behavior of different models,testing the the contents of 5-HT,5-HIAA,TPH2,MAO,IDO in rat raphe nucleus,to explore the mechanism of the effect of Simplified Chaihu Shugan Powder on the production and metabolism of 5-HT,in order to provide objective experimental evidence of the depression.Materials and methods:1.Forty SD rats were randomly divided into control group,model group,fluoxetine group,SCSP group.We established model by injecting reserpine,We treated them by fluoxetine and SCSP for 14 consecutive days at the same time.2.We recorded the general condition,behaviors after the experiment.The contents 5-HT and 5-HIAA in raphe nucleus were detected by High Performance Liquid Chromatography(HPLC-ECD).The contents of TPH2,MAO,IDO in raphe nucleus were detected by PCR.Results:1.Comparison of general situation of rats in each group:Comparing to control group,the model group,fluoxetine group and SCSP group showed different degrees of blepharoptosis,hair erection,back tension and weight loose in succession.Comparing to model group,the above situation has decreased influoxetine group and SCSP group.Comparing to control group,the body weight and weight gain of model group,fluoxetine group and SCSP group decreased significantly(P<0.01).Comparing to model group,the body weight and weight gain of fluoxetine group and SCSP group increased(P<0.05).There was no significant difference between SCSP group and fluoxetine group.2.Comparison of behaviors of rats in each group:Forced Swimming Test: Comparing to control group,the resting time in model group and SCSP group increased significantly(P<0.01),while that in fluoxetine group increased(P<0.05).Comparing to model group,the resting time in fluoxetine group and SCSP group decreased significantly(P<0.01).There was no significant difference between SCSP group and fluoxetine group.Open Field Test: Comparing to control group,the Total Distance and Zone Transition decreased significantly(P<0.01)in model group,the Total Distance has no significant difference but the Zone Transition decreased significantly(P<0.01)in fluoxetine group and SCSP group.Comparing to model group,the Total Distance increased significantly(P<0.01)but the Zone Transition has no significant difference in fluoxetine group and SCSP group.There was no significant difference between SCSP group and fluoxetine group.3.Comparison of the contents of TPH2 and 5-HT in raphe nucleus of rats in each group:Comparing to control group,the contents of TPH2 and 5-HT decreased significantly(P<0.01)in model group,but has no significant difference in fluoxetine group and SCSP group.Comparing to model group,the contents of TPH2 and 5-HT increased significantly(P<0.01)in fluoxetine group and SCSP group.There was no significant difference between SCSP group and fluoxetine group.4.Comparison of the contents of IDO in raphe nucleus of rats in each group:Comparing to control group,the contents of IDO in model group increased significantly(P<0.01),while that increased(P<0.05)in fluoxetine group,and there was no significant difference in SCSP group.Comparing to model group,the contents of IDO decreased significantly(P<0.01)in fluoxetine group and SCSP group.There was no significant difference between SCSP group and fluoxetine group.5.Comparison of the contents of MAO and 5-HIAA in raphe nucleus of rats in each group:MAO: Comparing to control group,the contents of MAO increased(P<0.05)in model group and SCSP group;there was no significant difference among other groups.5-HIAA: Comparing to control group,the contents of 5-HIAA decreased significantly(P<0.01)in model group and SCSP group;there was no significant difference in fluoxetine group.Comparing to model group,the contents of 5-HIAA increased significantly(P<0.01)in fluoxetine group but no significant difference in SCSP group.Comparing to fluoxetine group,the contents of 5-HIAA decreased significantly(P<0.01)in SCSP group.Conclusions:1.SCSP can improve the depressive-like behavior caused by reserpine.2.SCSP can increase the contents of 5-HT in raphe nucleus of depressive rats caused by reserpine.3.SCSP can increase the contents of 5-HT by increasing the contents of TPH2 and reducing the contents of IDO in raphe nucleus.
