Objective:To investigate the effect of high-fat diet on food intake and emotional activity in mice,and whether Chai hu Shu gan Powder,which function by regulating mood and relieving liver depression,could affect the level of emotion regulating factors in the central nervous system to regulate the expression of hypothalamic ingestion peptide and appitite and finally affect the feeding behavior.Research methods:1.After a week of adaptive feeding,4-week-old C57BL/6 male mice were randomized into the control group,the model group and the drug group.The control group feed normal,model group and drug group was given 60%fat feed two weeks,and then start drug intervention treatment,the normal group and model group mice to normal saline lavage at the same time,and drug group give the Chai hu Shu gan Powder.Feeding for 14 weeks,weekly monitoring of blood glucose and body weight.2.After 10 weeks,intraperitoneal glucose tolerance test(IPGTT)and intraperitoneal insulin tolerance test(IPITT)were performed.Small Animal Physiological Monitoring System(CLAMS)were applied to measure the energy metabolism of mice by recording food consumption,oxygen consumption,carbon dioxide production for continuous 24 hours.3.Mice behavior experiment including the open field experiment,suspended tail test,forced swimming test were performed to measure mood of mice in each group.4.The expression of NPY and POMC in the hypothalamus of each group was detected by real-time PCR.The level of hypothalamic feeding related proteins were detected by immunohistochemistry and Western blot.5.Serum triglycerides,cholesterol and monoamine neurotransmitters in hippocampus were determined by ELISA.Results:1?Body weight and blood glucose:Compared to the control group,the body weight and blood glucose were significantly increased in the HFD group.Chai hu Shu gan Powder group could reduce the body weight and blood glucose induced by high fat diet.2?IPGTT and IPITT experiments and Serum lipid levels showed that the insulin sensitivity and glucose tolerance were decreased in HFD group compared to control group.Triglycerides and Cholesterol levels in HFD group were higher than those in control group.Compare to the HFD group,Chai hu Shu gan Powder group showed a tendency to improve glucose tolerance but no efforts to insulin sensitivity.There were no difference between Chai hu Shu gan Powder group and HFD group in triglycerides and cholesterol levels.3?CLAMS data showed that the food consumption of mice in the HFD group was significantly lower than that of the control group.O2 consumption and CO2 release and energy dissipation were decreased;the intake of Chai hu Shu gan Powder Group was more than that of the HFD group,the O2 consumption,CO2 release and energy consumption were all higher than those in HFD group.4?Behavioral Results:1.Open-field test:Compared to the normal diet group,the total movement distance,peripheral movement distance and numbers of rearings of mice in HFD group were lower,the central area residence time in HFD group was increased;Compared to the HFD group,the total movement distance,peripheral movement distance and numbers of rearings of Chai hu Shu gan Powder group was increased.There was no difference between the Shugan Shugan Powder group and the HFD group in the central area residence time.2 Tail suspension test:I.Immobility time:The immobility time of HFD group increased compared with the control group.The Chaihu Shugan San group had a decreasing trend compared with the HFD group.II.High-strength struggle time:The HFD group had a shorter struggle time than the control group.The struggling time of Chaihu Shugan San group was longer than that of HFD group.3 Forced swimming experiment:I.Immobility time:The immobility time of HFD group increased compared with the control group.The Chaihu Shugan San group had a tendency to decrease compared with the HFD group.II.High-strength struggle time:The HFD group had a shorter struggle time than the control group.The struggling time of Chaihu Shugan San group was longer than that of HFD group.5?Ingestion of Peptide:The results of PCR showed that the levels of NPY mRNA and POMC mRNA in the HFD group were lower than those in the control group.The NPY mRNA and POMC mRNA levels in the Chaihu Shugan Powder group were higher than those in the HFD group.There was no statistical difference between the two groups;Western Blot,Immunohistochemistry showed that the expression of NPY and POMC protein in HFD group was lower than that in control group.The expression of NPY and POMC protein in Chaihu Shugan Powder group was higher than that in HFD group,but there was no statistical significance.6?Monoamine neurotransmitters in hippocampus:The 5-hydroxytryptamine,dopamine,and norepinephrine levels in the HFD group were lower than those in the control group.The levels of 5-hydroxytryptamine,dopamine,and norepinephrine in the Chaihu Shugan Powder group were higher than those in the HFD group,and the increase of serotonin was statistically significant.Conclusion:1?The long-term high-fat diet mice have a tendency of depression,andChai hu Shu gan Powder can increase the level of monoamine neurotransmitters in the hippocampus,especially increase the serotonin level to improve the bad mood,and have some antidepressant effect.2?The food intake of mice with long-term high-fat diet decreased.Chai hu Shu gan Powder can regulate the hypothalamus feeding peptides,which promotes mice's feeding effect to a certain extent,and can increase the metabolic rate of mice,and thus control the increase of body weight.
