| ObjectiveTo investigate the role of sophocarpine on proliferation,apoptosis and mitochondrial membrane potential of SMMC-7721 and LO2 cells.MethodsWe chose SMMC-7721 and LO2 cells for culturing and selected out two suitable concentrations of sophocarpine according to IC50.MTT,flow cytometry and mitochondrial membrane potential test were used to examine changes of biological behaviors of these two cells.In addition,western blot was also used to analyze the expression of Bcl-2,Bax and Cleaved-Caspase3.ResultsThe IC50 of the SMMC-7721 was 536.3(95% CI: 412.7-697.0).thus,we selected 500 mg/L and 1000mg/L to treat cells.The cell distribution was decreased and the cells were shrunk after treated with sophocarpine.The results of MTT showed that the OD values of both cells were significantly decreased with the increase of the concentration of sophocarpine(P<0.05).In the same concentration,there was no significant difference of proliferation between the two cells(P>0.05).The total apoptosis rate was increased as the concentration of sophocarpine was increased,but the apoptosis increasing rate of high concentration in LO2 was significantly higher than that in SMMC-7721(P<0.05).The mitochondrial membrane potential was also decreased with the increase of the concentration of sophocarpine(P<0.05).in addition,the levels of bcl-2 in these two cells were both decreased,while the levels of Bax and Cleaved-Caspase3 were increased,with the increase of sophocarpine.ConclusionsSophocarpine could suppressed the cell proliferation,decreased the mitochondrial membrane potential,and promoted the cell apoptosis of SMMC-7721 and LO2 through upregulating levels of Bax and Cleaved-Caspase3 and downregulating the level of Bcl-2,which implied that sophocarpine caused liver injury inevitably when killing liver carcinoma cells. |
