OBJECTIVES:This study was designed to explore the influence of hematoxylin on:①the inhibition ratio of hepatocellular carcinoma cell;②he adhesion of hepatocellular carcinoma cell Hep-G2;③the invasion of hepatocellular carcinoma cell Hep-G2;④he inhibition ratio of HUVEC;⑤. the angiogenesis of CAM.STUDY METHODS:Experimental part:①by using the MTT, this article tests the influence of hematoxylin with different concentrations on the inhibition ratio of hepatocellular carcinoma Bel-7402, Hep-G2, and the effect of time on the proliferation of drugs;②y using the MTT, this article tests the influence of hematoxylin with different concentrations on the adhesion of hepatocellular carcinoma Hep-G2, and the effect of time on the proliferation of drugs;③by using Transwell chamber model, this article observes the influence of hematoxylin on the invasion of hepatocellular carcinoma Hep-G2;④by using the MTT, this article tests the influence of hematoxylin with different concentrations on the inhibition ratio of HUVEC, and the effect of time on the proliferation of drugs;⑤this article observes the influence of hematoxylin on the angiogenesis of CAM by building a CAM model.THE RESULTS:①the influence of hematoxylin on the inhibition ratio of hepatocellular carcinoma Bel-7402、Hep-G2: When the concentration of hematoxylin is2p g/ml,there is no significant effect on inhibition ratio of hepatocellular carcinoma Bel-7402、Hep-G2. When the drug concentration is200μg/ml, the hematoxylin inhibits the proliferation of cell lines. The inhibition ratio rise to78.56%and87.38%. Compared with the control group, the hematoxylin has a significant effect on the inhibition of the proliferation of cell lines (P <0.015). The inhibition ratio is increasing with the increase of the concentration and time. It suggests that the inhibition ability of hematoxylin on the proliferation of cell line is time-dependent and dose-dependent.②the influence of hematoxylin on the adhesion of hepatocellular carcinoma Hep-G2:When the concentration is200μg/ml, the influence of hematoxylin on the adhesion of hepatocellular carcinoma becomes stronger when the time is longer. It affects79.33%of hepatocellular carcinoma at150min. Compared with the control group, the hematoxylin has a significant effect on affecting the adhesion of hepatocellular carcinoma (P<0.01). When the concentration is2μg/ml, the affecting of hematoxylin is weakened significantly. It suggests that the affection ability of hematoxylin on the inhibition ratio of the proliferation of cell line is time-dependent and dose-dependent③the influence of hematoxylin on the invasion of hepatocellular carcinoma Hep-G2:When the concentration is200μg/ml, hematoxylin can inhibit67.29%of hepatocellular carcinoma cell lines. Compared with the control group, the hematoxylin has a significant effect on the inhibition of the hepatocellular carcinoma (P<0.01). When the concentration of hematoxylin is20μg/ml and2μg/ml, the inhibition ratio is34.46%、14.18%respectively. This result shows that the influence of hematoxylin on inhibiting the invasion of hepatocellular carcinoma Hep-G2is declining with the decreasing of the concentration of hematoxylin.④he influence of hematoxylin on the inhibition ratio of HUVEC:When the concentration of hematoxylin is1μg/ml, there is no significant effect on inhibition ratio of HUVEC. When the concentration of hematoxylin is100μg/ml, the hematoxylin inhibits the proliferation of cell lines. The inhibition ratio rise to83.72%. Compared with the control group, the hematoxylin has a significant effect on the inhibition of the proliferation of cell lines (P<0.015). The influence of hematoxylin on HUVEC is time-dependent and dose-dependent.⑤he influence of hematoxylin on the angiogenesis of CAM:When the concentration is200μg/ml, hematoxylin has a strong inhibited effect on the angiogenesis of CAM. The inhibition ratio is100%. When the concentration of hematoxylin is20ug/ml and2ug/ml, the inhibition ratio is56.25%、37.5%respectively.CONCLUSION:①hematoxylin can lower the inhibition ratio of the hepatocellular carcinoma Bel-7402、Hep-G2;②hematoxylin is a inhibitor of the adhesion of hepatocellular carcinoma Hep-G2;③hematoxylin can control the invasion of hepatocellular carcinoma Hep-G2;④hematoxylin has a inhibited effect on HUVEC;⑤hematoxylin has a strong inhibitory effect on the angiogenesis of CAM.
