?Objective?The aim of the study was to determine KCaa channels expression in human villous trophoblasts and their role in proliferation,apoptosis and invasiveness of HTR-8/SVneo cells and human villous trophoblasts at fist term pregnancy;and ultimately to improve the pathogenesis of preeclampsia.?Methods?1 BKCa channels?subunit and?1 subunit,IKCaa channels,SKCaa channels expression in villous trophoblasts at fist term pregnancy was described using immunochemistry analyses.2 BKCa channels?subunit and?1 subunit,IKCaa channels,SKCaa channels expression in HTR-8/SVneo cells was described using cell culture immunofluorescence analyses.3 HTR-8/SVneo cells viability was measured by cell counting kit?cck-8?assay in the presence of NS1619(BKCa channels activitor),ibTX(BKCa channels blocker),NS309(IKCaa channels and SKCaa channels activitor),Apamin(SKCaa channels blocker)and TRAM34(IKCaa channels blocker).4 Apoptotic rate of HTR-8/SVneo cells was measured by flow cytometry analyses in the presence of NS1619,ibTX,NS309,Apamin and TRAM34.5 Invasive ability of HTR-8/SVneo cells was measured by transwell method in the presence of NS1619,ibTX,NS309,Apamin and TRAM34.?Results?1 BKCa channels?subunit and?1 subunit,IKCaa channels,SKCaa channels were expressed at cytomembrane and cytoplasm in villous trophoblasts at fist term pregnancy.2 BKCa channels?subunit and?1 subunit,IKCaa channels,SKCaa channels were expressed at cytomembrane and cytoplasm in HTR-8/SVneo cells.3 Activating BKCa channels,IKCaa channels,and SKCaa channels promoted HTR-8/SVneo cells proliferation;while blocking their function not brought obvious effection.4 Enhanced activities of BKCa channels was found to be correlated with increased apoptosis of HTR-8/SVneo cells,whereas enhanced of IKCaa channels,SKCaa channels and inhibition of BKCa channels,IKCaa channels,and SKCaa channels were not significantly affect apoptosis of HTR-8/SVneo cells.5 Enhanced activities of BKCa channels,IKCaa channels,and SKCaa channels were found to be correlated with inhibition invasivenessof HTR-8/SVneo cells,whereas blocking of BKCa channels,IKCaa channels,and SKCaa channels were not significantly affect invasiveness of HTR-8/SVneo cells.?Conclusion?The pressent results suggest that KCa channels are expressed in villous trophoblasts at fist term pregnancy.KCa channels play an important role in proliferation,apoptosis and invasiveness in HTR-8/SVneo cells.Taken together our results indicate that KCa channels may be considered an important target related to the action of villous trophoblasts at fist term pregnancy in spiral arteryremolding and may play a role in pathogenesis of preeclampsia.
