Unsaturated fatty acids(UFAs),such as the w-6 and w-3 fatty acids,which have multiple roles in membrane structure,lipid metabolism,blood clotting,blood pressure and have been linked to Cardiovascular diseases.Our previous research show that there is an accumulation of unsaturated fatty acids in the placenta tissue of preeclampsia[1].However,it is not yet clear how unsaturated fatty acids affect trophoblasts and preeclampsia.Therefore,we added various types of UFAs to Htr8 trophoblasts and villi explants to observe their effects on the cell and tissue level.Our research found that,with the addition of 100μM highly unsaturated fatty acids AA(20:4),EPA(20:5),DHA(22:6),the lipid peroxidation level of Htr8trophoblast cells increased,the migration and invasion ability was significantly weakened,and the proliferation There is no significant difference in apoptosis ability.Consistently,100μM highly unsaturated fatty acids AA(20:4),EPA(20:5),DHA(22:6)weakened the migration ability of villi explants.The high concentration of UFAs may cause lipid peroxidation in Htr8 cells,which affects the mitochondrial morphology and function of Htr8 cells,resulting in the reduction of ATP production of Htr8cells and the weakening of invasion and migration capabilities.It aggravates the trophoblast cell recasting disorder of the uterine spiral artery,leading to placental ischemia and hypoxia,thereby participating in the preeclampsia process. |
