Part oneChemical hypoxia affects trophoblasts biology behaviorsObjective: To reconstruct extravillous trophoblasts model in preeclampsia,to investigatethe changes of extravillous trophoblasts biology behaviors(proliferation, apoptosis,migration and invasion) in the condition of chemical hypoxia by cobalt dichloride (CoCl2)in the pathogenesis of preeclampsia.Methods:1.Using CoCl2to construct chemical hypoxia extravillous trophoblasts model.2.Using CCK8method、Flow cytometry、transwell methods to investigate the change ofproliferation、apoptosis、invasion and migration in extravillous trophoblasts.Results:1.Compared with control groups, the activity of extravillous trophoblasts was decreased inthe chemical hypoxia condition of6-72hours. And the activity of trophoblasts obviouslydecreased with the extension of incubation time, trophoblasts had the highest activity whenincubation for12hours.2.Compared with control groups, the total apoptosis rate of extravillous trophoblastssignificantly increased in the chemical hypoxia condition of12-72hours, trophoblasts hadmost pronounced total apoptosis rate increase when incubation for chemical hypoxia of48hours.3.Compared with control groups, the migration number of extravillous trophoblasts reducedin the chemical hypoxia condition of24hours.4.Compared with control groups, the invasion number of extravillous trophoblastsobviously reduced in the chemical hypoxia condition of48hours.Conclusion:Chemical hypoxia by CoCl2affects the behavior of trophoblasts biology. Using recombinant human maspin protein intervention trophoblasts under the conditionof normal or chemical hypoxia.3. Using CCK8method、Flow cytometry、transwell methods to investigate the change ofproliferation、apoptosis、invasion and migration in extravillous trophoblasts.Results:1.the activity and total apoptosis of extravillous trophoblasts had no change after givinghuman recombinant maspin protein under the condition of normal or chemical hypoxia.2.the migration and invasion number of extravillous trophoblasts obviously reduced aftergiving human recombinant maspin protein under the condition of normal or chemicalhypoxia.Conclusion:human recombinant maspin reduced the invasion and migration ability of extravilloustrophoblasts, but it has no obvious effect on the proliferation and apoptosis ability ofextravillous trophoblasts. So we speculated that maspin may restrain the ability ofmigration and invasion of trophoblasts, lead to shallow placenta implantation to participatein the pathogenesis of preeclampsia.
