At present,the pathogenesis of the chronic prostatitis of type ? is not fully understood,and the immunogenicity mechanism and theory of the oxidative stress reported in many research that account for the majority.Antibiotics treatment for this type prostatitis is effective,in which the recurrence rate has been more than high,otherwise some patients have poverty curative effect,thus find a different approach to investigate the pathogenesis and treatment of chronic prostatitis ? type is necessary.Recently,mounting number of literatures have been proposed the microbial imbalance has the high correlation with prostate disease,it suggested that changes in gut microorganisms likely one of the environmental factors of the development of the CP/CPPS.Additionally,it is also suggested that various plant polysaccharides can perform a crucial function in synergistic antiinflammation,regulating immunity and modulating the intestinal flora disorder,and some of the results are encouraging.In the development of prostatitis for type ?,the amount of oxygen free radicals increased in the body or the capacity of scavenging oxygen free radicals weakened,subsequently the body is moving into the state of oxidative stress.At the same time,the structure and function of the body will be damaged by peroxidation,which resulting in lipid peroxidation of cell proteins,DNA,cell membranes and organelles.So it might be considered that plant polysaccharides and hydrogen-rich water respectively can be a new treatment,by which we can evaluate the effect on chronic prostatitis of type ?,and try to reveal the pathogenesis of chronic prostatitis of type ? from the angle of gut microbiome.Objective: 1.To establish the model of the chronic prostatitis of type ? in rats,and evaluate the success rate of the establishment of the models.2.To evaluate the effect of plant polysaccharide on the chronic prostatitis of type ? in rats,and attempt to put forward the perspective that CP/CPPS give rise to microbiome disorder.3.From the Angle of oxidative stress of the body,to intervene the chronic prostatitis of type ? by using the hydrogen-rich water,and to observe the intervention effect on it,and to explore its application value.Methods: 1.Wistar rats were randomized into four groups of equal number to receive prostatic envelope injection of normal saline(NS control group),Freund's complete adjuvant(CFA group),and CFA with purified prostatic proteins at 15 mg /ml(LPP-CFA group)and CFA with purified prostatic proteins at 80 mg /ml(HPP-CFA group).At 7 days after modeling,the rats were sacrificed for hematoxylin-eosin(HE)staining of the prostate tissue and detection of the inflammatory factors IL-8,IL-10 and TNF-? in the serum.2.The model rats were divided into the normal control group(Control)and model group(MOD-1,MOD-2,MOD-3,MOD-4),which are by intragastric saline solution;Plant polysaccharide(PPS)or hydrogen-rich water(HRW)treatment group by intragastric polysaccharide liquor or hydrogen-rich water,and the group of the treatment in rats with CP/CPPS were continuously sacrificed within 1 week of gavage(1W group,PPS-1 / HRW-1),2 week of gavage(2W group,PPS-2 / HRW-2),3 weeks of gavage(3W group,PPS-3 / HRW-3),and 4 weeks of gavage(4W group,PPS-4 / HRW-4)for hematoxylin-eosin staining of the prostate tissue,for detection of the cytokines IL-6,IL-8,IL-10 and TNF-? in the serum,IL-1?,MDA,SOD in the prostate tissue homogenate,and for the quantitative analysis of TNF-? and tight junction protein(occludin protein)in the prostate tissue.Results: 1.Pathological examination exhibited morphological damage to the prostatic tissue and inflammatory cellular infiltration in the experimental rats.The serum level of IL-8,IL-10 and TNF-? was significantly higher in the CFA,LPP-CFA and HPP-CFA groups than in the control.2.In comparison to the normal control group,the rats in the other experimental groups had been treated for 4 weeks.The levels of IL-1?in the prostate tissue homogenate,and IL-6 and il-8 in serum were significantly decreased in each treatment group.Nevertheless the level of IL-10 in serum began to rise from week 1 and cease to do in week 2,succeeding gradually decreased in latter weeks.Furthermore the level of TNF-?in serum has a non-significant decreasing trend.The level of SOD each week after intervention,but MDA was the opposite.The expression of occludin protein of the rat's prostate gland epithelial was increased by week measured by immunohistochemistry method,as expected,the expression of TNF-? was down-regulated.Conclusion: 1.Both low and high concentrations of purified prostatic proteins with CFA can be used for the construction of autoimmune prostatitis models in rats,while high concentration is preferable for its advantages of high efficiency that can abbreviate the modeling time and inducement of more consistent manifestations of autoimmune prostatitis.2.This study is expected to recommend for the treatment of CP/CPPS.
