Objective: This research aimed to discuss the expression of neuronal nitric oxide synthase(nNOS)in duodenal bulb of the perinatal rat,as well as its influence on muscle layer thickness.Methods: Chosen 06 adult Wistar rats(SPF,no harmful bacteria levels),including 20 male rats and 40 female rats,all for 10 weeks.Putted them in cage,according to the ratio as female: male 2:1.On the next day divided them and take female rat vaginal smear to microscopy,found that the time of sperm was defined as the first day of pregnancy.Random grouped the Wistar rats,and divided them into treatment group and control group.The treatment group was chosen with L-NAME preparations,and set for three dose groups(low dose,middle dose and high dose,each group of 10 pregnant rats),lavaged the group of drugged pregnant rats and injected the mouse in the perinatal period.On the other side,to the 10 control pregnant mice,only used saline lavage and the rat injected medicine in the perinatal period,then collect weight daily data of the rat in the perinatal period.Finally stratified sampling according to the weight,and put to death respectively after 2,7 and 14 days of birth.Returned the specimen,embedded with sliced paraffin,detected after immunohistochemical decade and checked duodenal bulb type neuronal nitric oxide synthase(nNOS)distribution density,HE stained and measured the thickness of muscularis by duodenal bulb in microscope.Results:(1)The weight increase of the drug group rat in the first week of perinatal period was lower than the control group,and was the same with the control group in the second week.No statistical significance within the drug used group.(2)Compared the treatment group rat in the perinatal period of nNOS distribution density with control group,the expression of nNOS quantity did not change on 7-day-age,but increased on 14-days-age.(3)On the aspect of duodenal bulb muscle the layer thickness in the perinatal period,compared the drug group rat(7-day-age and 14-days-age)with the control group,there was no statistical significance.Conclusion: L-NAME can induce the increasing of the activity and expression of the newborn mice duodenal bulb muscularis nNOS,while keeping the layer thickness of the duodenal bulb muscle without affected.
