Abstract/Summary:
1ObjectiveIn the present study, we further focus on the anti-morphine psychic dependence effects of [Glu3,4,7,10,14]-Conantokin-G(Glu-Con-G) and [Glu3,4,7,10,14]-Conantokin-G [1-13](Glu-Con-G[1-13]) in male and femal mice. Western blotting was used to testify the changes of the levels of nNOS in hippocampus (Hp) of morphine-addicted mice to discuss its role in morphine addiction.2Experimental methodsMale&female Kunming mice (18-20g) were used in this experiment. The effect of morphine dependence is evaluated by conditioned place preference (CPP) model; the Western blotting was used to study the molecular changes.3ResultsThe results of behavioral experiment (CPP) show that, Glu-Con-G and Glu-Con-G [1-13](120,240,480pmol) by single intracerebroventricular administration inhibited the expression and reinstatement of morphine-induced CPP in male groups; Glu-Con-G [1-13](120,480pmol) by single intracerebroventricular administration inhibited the expression of morphine-induced CPP in female group.Both Glu-Con-G and Glu-Con-G [1-13](120,240,480pmol) by single intracerebroventricular administration had no invisible behavioral disorder under the experiment conditions.The results of Western blotting showed that, a certain dose of Glu-Con-G and Glu-Con-G [1-13] by single intracerebroventricular administration can attenuate the level of nNOS in hippocampus of morphine addictive mice.4Conclusions1. Single intracerebroventricular administration Glu-Con-G (120,240,480pmol) could attenuate the expression and reinstatement phases of morphine-induced CPP in male mice, but not in the female group.2. Single intracerebroventricular administration Glu-Con-G [1-13](120,240,480pmol) could attenuate the expression and reinstatement phases of morphine-induced CPP in male mice, and attenuatethe expression phases of morphine-induced CPP in female at the doses of120and480pmol.3. Single intracerebroventricular administration of both Glu-Con-G and Glu-Con-G [1-13](120,240,480pmol) cause no invisible disorder of locomotor activity or exploratory behavior under our experiment conditions.4. The levels of nNOS were increased in hippocampus of morphine addictive male mice, and could be attenuated by Glu-Con-G and Glu-Con-G [1-13] at the doses of120,240,480pmol.
