Rationales:Aging is an inevitable trend in the development of living organisms.Aging is usually accompanied by senile degenerative diseases such as Alzheimer's disease(AD),amyotrophic lateral sclerosis(ALS),Parkinson's disease(PD)and heart disease.Its complexity is affected by many factors.With the aging,the diseases related to aging and aging have seriously affected the health of the elderly.Therefore,how to prevent aging and improve the quality of life and health of the elderly has become one of the major problems in the medical field.Astragalus is the dry root of the leguminous plants Astragalus membranaceus(Fisch.)Bge.Var.Mongholicus(Bge.)Hsiao and Astragalus membranaceus(Fisch.)Bge.Astragaloside ? is a representative component of astragaloside saponins in Radix Astragali.Astragaloside has several pharmacological activities,including antibacterial,Anti-fibrosis,anti-oxidant,antiinflammatory and neuroprotective effects.Studies have also shown that Water extract of Astragalus can extend the lifespan of zebrafish.In addition,some studies have shown that some components of Water extract of Astragalus also have antiaging effects.For example,Astragalus polysaccharides could prolong the lifespan of mice by scavenging reactive oxygen species,inhibiting mitochondrial permeability transition and increasing antioxidant enzyme activity,which indicated that Astragalus polysaccharides have antiaging effects.Astragalus saponins can delay the aging caused by D-galactose.The total flavonoids of Astragalus membranaceus can delay the acute aging of mice induced by D-galactose through increasing the non-specific immunity of mice.However,the antiaging effects of Astragaloside ? have rarely been documented,and antiaging mechanisms have not been systematically studied.In this paper,we conducted a series of studies to illustrate the effect mechanism of Astragaloside ? on antiaging in Caenorhabditis elegans.Objective: To study the effects of Astragaloside ? on the lifespan of Caenorhabditis elegans,and to explore the antiaging effects and mechanisms of Astragaloside ?.Methods: The effects of Astragaloside ? on the lifespan of nematodes were tested by using natural aging model,paraquat injury model and heat stress model.We also examined the effects of astragaloside on nematode motility.At the same time,in order topredict the anti-aging mechanism of astragaloside,NMR metabonomics was used to evaluate the changes of nematode endogenous metabolites.Finally,the mechanism of anti-aging effects of Astragaloside ? can be further elucidated through biochemical and genetic levels.Results: Astragaloside ? could prolong the lifespan of nematodes in natural aging model,paraquat injury model and heat stress model.The underlying mechanism may involveincreasing the activities of superoxide dismutase and catalase in the nematode,and inducing the mRNA expressions of SOD and CAT genes.Conclusion: Astragaloside ? can prolong the lifespan of nematodes and its mechanism of action is associated with antioxidation.
