Objective:To consult the relevant literature,sort out the origin and meaning of "Shen" and "Zhi" and the relevant theories of "Huangdi Nei Jing" from the theory of traditional Chinese medicine,sort out the understanding of AD in TCM from the point of view of disease name,etiology and pathogenesis,and discuss the rationality of the method of Anshen Yizhi in the treatment of AD combined with the treatment and prescription analysis of the method of Anshen Yizhi.The network pharmacology platform was used to analyze the possible targets and mechanisms of Anshen Yizhi method on AD,and related experiments were carried out on these targets and mechanisms with the application of model nematode(wild type N2 and AD transgenic model CL4176).To observe the effect of Anshen Yizhi method on c.elegans,and to explore the possible mechanism of Anshen Yizhi method in the prevention and treatment of AD.Methods:1.Theoretical discussion: collect and sort out the relevant literature,sort out the origin,meaning and related theoretical explanation of "Shen","Zhi" and AD in traditional Chinese medicine,analyze the treatment methods and prescriptions of the Anshen Yizhi method,and explore the theoretical basis of the Anshen Yizhi method in the treatment of AD.2.Experimental study: the effect of Suanzaoren decoction on AD was studied by using common wild type beautiful nematode N2 and AD model(A ? transgenic)nematode CL4176 as animal model.In order to observe the effect of Suanzaoren decoction on against A ? Protetoxicity and antioxidant level,the paralysis remission experiment and in vivo ROS level of A ?transgenic c.elegans model CL4176 were measured.The experiment of oxidative stress and life span of wild type nematode N2 was carried out to observe whether Suanzaoren decoction could improve its survival rate under oxidative stress and its ability to prolong the life span and delay senescence of c.elegans.At the same time,the contents of SOD,CAT and MDA were determined to observe the antioxidant activity of Suanzaoren decoction.Finally,real-time PCR was used to detect the expression of related genes pointed to by the results of network pharmacology.Results:1.Results of network pharmacology: according to the results of PPI and GO analysis of network pharmacology,we found that many targets and biological processes were related to antioxidation.2.The results of oxidative stress,ROS level,enzyme activity and MDA :content test showed that the survival rate of N2 nematode treated with Suanzaoren decoction was significantly increased under paraquat oxidative stress,while the ROS level of CL4176 nematode was decreased,the activities of antioxidant enzymes SOD and CAT in CL4176 nematode were increased,the content of MDA was decreased,and the activity of antioxidant enzyme SOD of N2 nematode was increased,but the activities of CAT and MDA did not change significantly.3.The results of paralysis remission experiment showed that Suanzaoren decoction could delay the paralysis process of CL4176 nematode and inhibit A ? aggregation toxicity,and its effective rate increased with the increase of concentration.4.Longevity test results: Suanzaoren decoction 1mg/m L and2mg/m L groups prolonged the life span of wild nematode N2,but its effect was not obvious.5.Real-time PCR results: Suanzaoren decoction significantly up-regulated the m RNA expression levels of sod-3,daf-16,hsp-16.2 and akt-1 genes in AD nematode.Conclusion:1.In the ancient books of traditional Chinese medicine,"Shen" and "Zhi" are closely related to AD,and they are senseless,and the misuse of divine opportunity can easily lead to the Anshen Yizhi method of AD.It may be an effective treatment for prevention and treatment of AD.2.The results of network pharmacology show that Suanzaoren decoction may act on AD by regulating the level of antioxidation.3.Suanzaoren decoction could improve the survival rate and SOD activity of N2 nematode under oxidative stress,but there was no significant change in CAT and MDA activity.At the same time,it can reduce the level of ROS in CL4176 nematodes,increase the activities of antioxidant enzymes SOD and CAT,and reduce the content of MDA,indicating that Suanzaoren decoction does have antioxidant effect,and the antioxidant effect in transgenic AD nematodes is more obvious than that in wild-type nematodes.4.Suanzaoren decoction can delay the paralysis process of CL4176 nematodes,inhibit A ? aggregation toxicity,and up-regulate sod-3,daf-16,hsp-16.2 and akt-1 motifs of CL4176 nematodes,suggesting that Suanzaoren decoction can improve the antioxidant capacity of beautiful nematodes and alleviate A ? toxicity.
