Objective: To investigate the differences in serum folic acid concentration and the methylation of PAX1 gene in peripheral leukocyte between oral squamous cell carcinoma patients,oral epithelial dysplasia patients and healthy volunteers.To explore whether there wass a correlation between serum folic acid concentration and the methylation of PAX1 gene in peripheral leukocyte,and if there was a correlation between these two levels and the clinical and pathological factors in patients with oral squamous cell carcinoma,and to expand new ideas for the prevention,early diagnosis and related treatment of oral cancer.Methods: Patients with oral squamous cell carcinoma and oral epithelial dysplasia were selected as the experimental group,and healthy volunteers as a control group.Collection of fresh blood samples from patients with oral squamous cell carcinoma diagnosed as oral squamous cell carcinoma in the First People's Hospital of Xiangtan City from June 2017 to August 2018.14 cases of fresh blood samples before treatment,27 cases of healthy volunteers who participated in the health examination of the First People's Hospital of Xiangtan City.All blood samples were taken from fasting peripheral venous blood in the early morning.Serum folic acid concentrations were determined by electrochemical luminescence immunoassay(ECLI)and the methylation status of CpG island in PAX1 gene promoter region of peripheral leukocyte was determined by real-time quantitative methylation specific PCR.SPSS22.0 software was used for statistical analysis of the related data.Results:1.Serum folic acid concentration in patients with oral squamous cell carcinoma and patients with oral epithelial dysplasia(22.69±8.93,27.07±10.69)was lower than the healthy control group(32.35±9.54),(P<0.05).2.The serum folic acid concentration in oral squamous cell carcinoma patients was negatively correlated with age(r=-0.096,P<0.05)and tumor size(r=-0.170,P<0.01).3.The PAX1 gene in peripheral leukocyte of the patients with oral squamous cell carcinoma,the oral epithelial dysplasia group and the healthy control group was hypomethylated.Conclusion:1.Folic acid was associated with the development of oral atypical hyperplasia and oral squamous cell carcinoma,and was of great significance for the prevention,early diagnosis and related treatment of oral cancer.2.There was no correlation between serum folic acid and the methylation of PAX1 gene in peripheral leukocyte in oral squamous cell carcinoma;and it was not considered that the occurrence of oral squamous cell carcinoma is associated with the methylation of PAX1 gene in peripheral leukocyte.
