Study On The Mechanism Of Xier Zidian Zhenxiao Granule (Non-decocted) On Vascular Inflammation In Allergic Purpura Rabbit Model

Objective:Through experimental study of Zidian Zhenxiao on coagulation routine.D-dimer.IL-6;to explore the role of Zidian Zhenxiao in the treatment of Henoch-Schonlein purpura and its mechanism;provide a scientific basis for the preclinical study of Zidian Zhenxiao.Method:1.Experimental white rabbits were randomly divided into 5 groups:blank group,model group,Zidian Zhenxiao of high,medium and low dose group.2.Model Preparation: The ginger,pepper,emu in accordance with the ratio of1: 1: 1 boiling,dubbed 15 g / 100 ml of water solution,except for the blank group,the remaining groups were administered on the 1st 1 intragastric administration1 ml,The blood heat constitution model was established 3 weeks after continuous administration;0.5ml of 0.9% sodium chloride injection was added with 10 mg of ovalbumin(F5505,SIGMA)and complete Freund’s adjuvant(SIGMA)according to1: 1 Proportion mixed,with 1ml mixed emulsion solution,the rabbits were injected intraperitoneally once a week,uninterrupted injection for 3 weeks to build HSP rabbit model.The blood samples were collected from the ear vein,and the routine coagulation tests were performed.The DD2,CRP and IL-6 values were measured and statistically analyzed to determine whether the model preparation was successful.4.Administration: blank group,model group under the same feeding conditions to give saline;high,medium and low dose group given Zidian Zhenxiao gavage administration.After continuous administration for 10 days,blood was collected from the ear vein to detect coagulation routine,DD2.CRP and IL-6 values,and observe the effect of the Zidian Zhenxiao.Result:In this animal experiment,after successful preparation of HSP model,the values of PT.APTT.CRP.D-D2 and IL-6 in the model group and the drug group were significantly higher than those in the blank control group,respectively,Analysis,with statistical significance.In the high,medium and low dose groups were compared with the blank control group 10 days after the administration of the Zidian Zhenxiao,the results showed that the high,medium and low dose groupshad better curative effect,and the middle dose group had higher effect on PT.APTT.CRP.D-D2.IL-6 value of the most obvious effect.The low dose group followed by the high dose group again.All have statistical significance.It can be inferred that Zidian Zhenxiao can effectively control the onset of HSP and can also prevent the occurrence of kidney damage,and can reduce the degree of kidney damage.Conclusion:1.Zidian Zhenxiao have a remarkable curative effect on ecchymoses of HSP rabbit model;2.The Zidian Zhenxiao coagulation routine,DD2,CRP and IL-6 values in HSP rabbit model tend to be normal,and the Zidian Zhenxiao are proved to be effective in terms of coagulation mechanism and inflammatory factors And scientific.
