Objective: The aim of this study is to investigate the inhibitory effect of Xiao’er Zidian Zhenxiao Granules(XZKL)on the inflammation of human umbilical vein endothelial cells(HUVECs)via NF-κB signaling pathway.Meanwhile,it was confirmed that XZKL could inhibit the formation of neutrophil extracellular trap(NETs),thus reducing vascular inflammatory injury.Methods: LPS-induced HUVECs were used as an inflammation model and different concentrations of XZKL were added for intervention,and the optimal drug concentration was screened by CCK-8 method.We detected the expression of proteins associated with NF-κB inflammatory signaling pathway,such as TLR4,My D88,IKBKB,p-Ik B-α,Ik B-α,NF-κB,p-p65,IL-1β and TNF-α by Western blot and the levels of TNF-α inflammatory factors using enzyme-linked immunosorbent assay(ELISA),followed by flow cytometry to detect the levels of reactive oxygen species(ROS)in HUVECs.In an all-trans retinoic acid(ATRA)-stimulated HL-60 cell induction into neutrophil model,the effect of XZKL on the associated proteins of NETs was detected by Western blot after 4 h of induction with the addition of PMA.Results: In the LPS-induced inflammation model of HUVECs,XZKL reduced the expression levels of TLR4,My D88,IKBKB,p-Ik B-α,p-p65,IL-1β and TNF-α proteins,while Ik B-α protein expression was increased.XZKL effectively inhibited the release of the cellular inflammatory factor TNF-α and reduced ROS production in HUVECs.XZKL could reduce the formation of NETs and decrease the inflammatory response.Conclusion: XZKL inhibits LPS-induced vascular inflammation by suppressing NF-κB signaling pathway,and also inhibits the formation of NETs,thus reducing vascular injury.
