Objective:The aim of this study was to explore the effect of technetium methylenediphosphonate(99Tc-MDP)on RAW264.7 and MC3T3-E1 cells,and provide the theoretical basis for the clinical treatment of bone resorption diseases related to osteoclasts such as ameloblastoma of the jaws.Methods:Different concentrations of 99Tc-MDP were used to treat mononuclear macrophage leukemia cells(RAW264.7)and mouse embryonic osteoblast precursor cells(MC3T3-E1).The cell proliferation inhibition rate was analyzed by CCK-8.Cell apoptosis and cell cycle were evaluated by flow cytometry.Immunocytochemistry and Western blot were used to detect the expression of RANKL,OPG and RANK protein.Results:The inhibitory rates of RAW264.7 cells treated with 0.01 ?g/ml and 100 ?g/ml of 99Tc-MDP for 48 h were 6.09±2.38%and 52.57±8.38%,respectively.The inhibition of RAW264.7 cells by 99Tc-MDP with different concentrations was detected.The effect was significantly stronger than the control(P<0.05),and induced apoptosis and decreased the expression of RANK protein.The survival rates of MC3T3-E1 cells treated with 99Tc-MDP at 0.01 ?g/ml and 100?g/ml for 48 hours were 123.08 ± 2.87%and 76.64 ± 4.29%,respectively.Compared with the control group,the survival rates of MC3T3-E1 cells treated with 0.01?g/ml of 99Tc-MDP significantly increased(P<0.05),promoted MC3T3-E1 cells proliferation,and increased the expression of OPG and RANKL proteins.The high concentration of 100?g/ml of 99Tc-MDP significantly inhibited MC3T3-E1 cells and decreased the expression of OPG and RANKL protein.Different concentrations of zoledronic acid(ZOL)significantly inhibited RAW264.7 cells and MC3T3-E1 cells than control(P<0.05).Conclusion:99Tc-MDP can inhibit RAW264.7 cells,increase as the concentration and duration of action,induce apoptosis and inhibit the expression of RANK protein.The effect of 0.01 ?g/ml of low concentration of 99Tc-MDP can promote the proliferation of MC3T3-E1 cells and increase the expression of OPG and RANKL protein.ZOL had an inhibitory effect on RAW264.7 and MC3T3-E1 cells,and increased as the increase concentration and duration of action.99Tc-MDP may have a certain therapeutic effect on jaw ameloblastoma.
