| Objective:To observe the treatment of the 99Tc-MDP on adjuvant arthritis(AA) in rat, and the influence on the PGE1 and PGE2 in serm, and the expression in aynovial. To explore the mechanisms of the 99Tc-MDP in the treatment of rheumatoid arthritis.Methods:In this study,30 Wistar rats were ramdomly assigned to three groups with 10 rats in each group:group A was normal control group,the adjuvant arthritis was induced in the rats by a sigle intradermal injection of 0.1ml normal saline into the subplantar area of the left hind paws, the corresponding volume of normal saline(2.5ml/kg) was given through caudal vein injection on the 10th day after the arthritis was induced every day, total 15 days; group B was AA group, the the adjuvant arthritis was induced in the rats by a sigle intradermal injection of 0.1ml Complete Freund Adjuvant into the subplantar area of the left hind paws, the corresponding volume of normal saline(2.5ml/kg) was given through caudal vein injection on the 10th day after the arthritis was induced every day, total 15 days; group C was 99Tc-MDP group, the the adjuvant arthritis was induced in the rats by a sigle intradermal injection of 0.1ml Complete Freund Adjuvant into the subplantar area of the left hind paws, the corresponding volume of 99Tc-MDP (2.5ml/kg) was given through caudal vein injection on the 10th day after the arthritis was induced every day, total 15 days. The left to right diameter of the left hind ankle, arthritis index were tested at different time points. Kill the rats by every group and blood sampling were centrifugal and frozen to use. The level of PGE1 and PGE2 in serum was measured by ELISA. The genual synovial tissues were acpuried and HE staining was done to observe the pathologic change by light microscope. Use spss 15.0 statistical software to analyse the above-mentioned data.Results:1. The effect of 99Tc-MDP on arthritis index of joint:At the beginning of the therapy, the arthritis index of B group were not significantly different from C group(P＞0.05); On the 2th week after therapy, arthritis index of B group became higher than those of C group(P＜0.05); When it came to the 4th and 6th week after therapy, arthritis index of B group becam more higher than those than C group(P＜0.01).2.The effect of 99Tc-MDP on the left to right diameter of the left hind ankle:At the beginning of the therapy, the left to right diameter of the left hind ankle of B group were not significantly different from C group(P＞0.05), but the left to right diameter of the left hind ankle of B group and C group were higher than A group; On the 2th week after therapy, the left to right diameter of the left hind ankle of B group became higher than those of C group(P＜0.05); When it came to the 4th and 6th week after therapy, the left to right diameter of the left hind ankle of B group became more higher than those than C group(P＜0.01); But the left to right diameter of the left hind ankle of A group were not significantly different from C group(P>0.05).3.The effect of 99Tc-MDP on the serum level of PGE1 and PGE2:On the 6th week after therapy, the level of PGE2 of B group and C group became higher than A group(P＜0.01); the level of PGE2 of C group became lower than B group; the level of PGE1 among A group,B group and C group were not significantly different from other group(P＞0.05).4. The pathology examinationv of rats left ankle in group B and C showed synovial hyperplasia and inflammatory cells, but the infiltration in treatment group C is less than group B. The synovial hyperplasia degree in group C is lower than group B.Conclusions:1.99Tc-MDP could significantly reduce inflammation, arthritis index and the left to right diameter of the left hind ankle of adjuvant arthritis rats.2.99Tc-MDP could reduce the level of PGE2 in blood plasma of adjuvant arthritis.3.99Tc-MDP had conspicuous anti-inflammatory effects on adjuvant arthritis in rats.
