| Objective: By copying the model of alcoholic liver fibrosis in rats,observe the li ver fat soluble particles on alcoholic liver fibrosis animal serum related index(Tbil,D bil,ALT,AST,GGT,ALP,ALB,GLOB,A/G)and hepatic fibrosis index(HA,LN,IV-C,PCIII)and liver tissues.To investigate the intervention effect of liver fat solubl e particles on alcoholic liver fibrosis,clinical expansion of liver fat soluble particles i ndications for prevention and treatment of traditional Chinese medicine and provide a theoretical basis for the development of disease,Chinese medicine research on the dise ase.Method: Sixty healthy SD rats,half male and half male,weight 200-220 g,were randomly divided into blank group(n = 8)and model group(n = 12)after one week of adaptive feeding.10 rats in each group were treated with alcohol and corn oil,wi th 10 rats in the control group with low dose of 1.0 g / kg of lipopolysaccharide,2.0g / kg of middle dose,4.0 g / kg of high dose of Ganshuang granule and 0.3g / kg of compound Ganshuang granule.The animal model of alcoholic hepatic fibrosis was established by intragastric administration of pyrazole.After 12 weeks of continuous a dministration,the model was successfully established.The normal group and the mode l group were given the same volume of normal saline for 6 weeks.During the course of oral administration of the drug,the rats continued to model the drug.At the end of the 12 th week,the rats fasted for 12 hours,and the next morning,the body was t aken out for the last time to weigh the rats.10% chloral hydrate was used to anesthe tize,abdominal aorta blood was preserved and centrifugation was used to prepare the serum.The serum was separated,and the related indexes of liver function were detect ed by automatic biochemical analyzer.The liver function was detected by automatic bi ochemical analyzer,and the liver fiber was determined by chemiluminescence method.The liver fiber was determined by chemiluminescence method.I).Liver tissue was fix ed in 10% neutral formalin,then dehydrated,embedded in paraffin,sectioned,stained with hematoxylin and eosin hehe).The pathological changes of liver and hepatic fibro sis were observed under microscope.Result:(1)After treatment,the conditions of rats of the Ganzhirong particles high,middle,and delow dose groups as wellas the Ganshuang granule group were significan tly improved.As compared to thecontrol group,the ALT AST GGT ALP levels in the serum of the model group increase dgreatly(P<0.01);the HA,LN,PIIINP,and IVC levels were also significantly higher(P<0.01);TPGLOBA/G level were also significan tly higher(P<0.05orP<0.01)ALB(P>0.05).(2)The liver cells in each group were observed under light microscope as follow s:In the normal group,the hepatocytes were arranged clearly,the nucleus was large and round,the hepatic cord was arranged radially in the central vein,the hepatic lob ule structure was clear,the hepatic sinusoids were normal,and the hepatic sinusoids were not destroyed.In the model group,there was no fatty degeneration of liver tissue,a large numb er of liver cells appeared vacuolation,and a little,focal necrosis with inflammatory ce ll infiltration,the degree was more serious,partial bridging necrosis,there was a large number of fibrous tissue deposition around the catchment area.Part of hepatic lobule structure was destroyed;Drug intervention group: compared with the model group,the necrosis of hepatoc ytes,infiltration of inflammatory cells and deposition of fibrous tissue in the catchmen t area were decreased in different dosage groups.Conclusion:After 12 weeks of oral administration of alcohol,corn oil and pyrazol e,the rat model of alcoholic liver fibrosis was successfully established.Lipopolysin gr anules could reduce bilirubin,protect hepatocytes and bile duct cells,regulate immunit y and resist fibrosis. |
