Background and object:Colorectal cancer(CRC)is a kind of disease whose morbidity and mortality is high in worldwide.It is the fifth most common cancer in the world,and third cancer-related death out in global.Its five-year survival rate is less than 65%.Once diagnosed,the disease was almost at the late stage,which brought a lot of restrictions to treatment.Studies have shown that 25% of patients have transferred when diagnosed.Early diagnosis of the disease has a great value to improve patients‘ life quality.Now it is urgently needed a tumor marker with high sensitivity and specificity to achieve early detection,early diagnosis and early treatment of colorectal cancer.Through two-dimensional chromatography and mass spectrometry technique in proteomics,we discover the differences between the cancer tissue and the corresponding tissue adjacent to carcinoma in the protein expression,and build difference protein expression spectrum of colorectal cancer.At the same time,we expect to select a tumor marker which has a high value in diagnosis and treatment of colorectal cancer.Method:This research mainly divided into the following sections: 1.Two-dimensional chromatography and mass spectrometry technique to build difference protein expression spectrum:1)Select colorectal adenocarcinoma tissue and its corresponding adjacent normal tissue,and extract the tissue proteins,then prepare peptide mixture.2)Two-dimensional chromatography and mass spectrometry technique to test difference protein expression.3)Analyze and build difference protein expression spectrum.2.Verify ATP5A1 expression in colorectal cancer: Select 3 cases of colorectal adenocarcinoma tissue and its corresponding adjacent normal tissue,and use Western Blot method to detect the target protein ATP5A1 expressed in colon cancer tissue and tissue adjacent to carcinoma.3.The relationship between the expression of ATP5A1 and clinical pathological parameters: 1)From the patient's age,sex,TNM stage,and presence of distant metastasis,etc.,to establish the clinical database.2)Using immunohistochemical method to detect ATP5A1 expression in colorectal cancer,and analyze the relationship between its expression and the patients with clinical pathological parameters.Results:1.Build difference protein expression spectrum: Analyze the different expressed proteins between the carcinoma tissue and adjacent to carcinoma tissues,get 25 different expressed proteins,of which 18 protein expression level was up-regulated and 7 protein expression was down-regulated in cancer tissues.2.Western Blot results showed that the expression of ATP5A1 in colon cancer tissues significantly higher than the amount of tissue adjacent to carcinoma.3.? Immunohistochemical results show that ATP5A1 is highly expressed in colorectal cancer,and main expressed in the cytoplasm and cell membrane.? Of 20 cases of colorectal cancer tissue,7 cases which ATP5A1 protein expression was positive.And of 20 cases of the corresponding tissue adjacent to carcinoma,none cases ATP5A1 protein expression was positive.ATP5A1 protein expression in colorectal cancer tissue is much greater than the corresponding tissue adjacent to carcinoma raised obviously,the difference was statistically significant(p<0.05).? ATP5A1 protein expression relate to TNM staging,the presence of distant metastasis(p< 0.05),and is highly correlated to the level of CEA.But has nothing to do with the patient's age,gender,diagnosis,and differentiation degree of colorectal cancer(p > 0.05).Conclusion:1.The expression of ATP5A1 in colon cancer tissues is significantly higher than the amount of tissue adjacent to carcinoma.2.ATP5A1 protein expression relates to TNM staging,the presence of distant metastasis,and is highly correlated to the level of CEA.But has nothing to do with the patient's age,gender,diagnosis and differentiation degree of colorectal cancer.3.ATP5A1 plays an important role in colorectal cancer occurrence and development,and has certain value for colorectal malignant degree,which laid a foundation for further study.
