Objective:To choose the "Dachangshu","Tianshu","Zusanli" and "Shangjuxu" four specific points,from the morphological point of view,to observe the therapeutic effects of electroacupuncture acupoints on four different models of acute and chronic colitis rats,compare the difference;from the perspective of molecular biology,the observation of Electroacupuncture four different acupoints on colonic myeloperoxidase rat model of acute and chronic colitis(MPO),serum IL-6,serum IL-10 content,to explore the protective mechanism of acupoints on colonic mucosal damage,provide laboratory basis for reasonable selection of acupoints for clinical treatment of two types of colitis.Methods:Experiment one: 60 rats were randomly divided into blank group,model group,Dachangshu,Tianshu,Zusanli and Shangjuxu group group,10 rats in each group.To establish acute colitis rat model with 6% acetic acid enema.After successful modeling,the points of rats to fix.In the model immediately,24 h,48h in the EA 1 times,30 minutes each time,the blank group and the model group rats were bundled fixed EA.The end of the third after treatment,rats were fasted,the next day with 10% chloral hydrate anesthesia respectively.Abdominal aortic blood centrifugation supernatant,detect serum in IL-6,IL-10 Content;rats anus and above 12 cm intestinal tissue,observation of colonic mucosal damage;after that,some lesions were taken from colon tissue to measure myeloperoxidase(MPO);In addition,the pathological changes of another colon tissue were observed.Experiment two: 120 male rats were randomly divided into blank group,model group,Dachangshu group.Tianshu,Zusanli group and Shangjuxu group,20 rats in each group.The rat model of chronic colitis by stimulating the immune system.The method of local acupoints electroacupuncture in rats with experimental methods,namely daily acupuncture 1 times,30 minutes each time,the blank group and the model group rats were bundled fixed.After 3 days of continuous electroacupuncture,10 rats in each group were killed randomly,and the rest rats were killed after 7 days of electroacupuncture.The method and index of observation were the same as experiment one.Results:1.From morphological observation,we can get:In the acute and chronic colitis experiments,the index of colonic mucosa damage in the model group was higher than that in the blank group,the difference was statistically significant(P < 0.01);Compared with the2.model group,the indexes of colonic mucosa damage in each acupoint group were decreased,and the difference was statistically significant(P < 0.01);Compared with the 3 day after electroacupuncture,there was no significant difference in the index of colonic mucosa between the acupoints of acute and chronic colitis(P > 0.05);Electroacupuncture of chronic colitis for 10 days,compared with the Tianshu group,Dachangshu group and Zusanli group colon mucosa damage index is high,the difference was statistically significant(P < 0.05 or P < 0.01),Shangjuxu group comparison,no statistically significant difference(P > 0.05).Except at group,comparing to the other three points group,the difference was not statistically significant(P > 0.05).3.From the analysis of the mechanism of action:In the acute and chronic colitis experiments,the content of MPO and serum IL-6 in the colon tissue of the model group were higher than that in the blank group,and the serum IL-10 content decreased,the difference was statistically significant(P < 0.01);Compared with the model group,the MPO content and serum IL-6 content in the colon tissue of each acupoint group decreased,and the serum IL-10 content increased,the difference was statistically significant(P < 0.05,or P < 0.01);Acute colitis electroacupuncture for 3 days,Tianshu group compared with the MPO content of other acupoints group colon tissue decreased,the difference was statistically significant(P < 0.05),each group on the content of serum IL-6,serum IL-10 were compared,the difference was not statistically significant(P > 0.05).Electroacupuncture of chronic colitis for 3 days,Tianshu group than Dachangshu,Zusanli decreased the MPO levels of the two groups,the difference was statistically significant(P < 0.05);there was no significant difference compared with group(P > 0.05);Shangjuxu group than Dachangshu,Zusanli decreased the MPO levels of the two groups,the difference was statistically significant(P < 0.05).The points between the groups of serum IL-6,serum IL-10 content,there was no statistically significant difference(P > 0.05).At 10 days,MPO content was higher than the other Tianshu acupoints group colon tissue decreased(P < 0.01).Compared with the Tianshu group,Dachangshu group serum IL-6 content is high,the difference was statistically significant(P < 0.05),Zusanli group,compared with the Shangjuxu group the content of IL-6 in serum,the difference was statistically significant(P > 0.05).Compared with the Tianshu group,low Dachangshu group the content of IL-10 in serum,the difference was statistically significant(P < 0.05),4.compared with the Zusanli group,Shangjuxu group of low serum IL-10 content,but the difference was not statistically significant(P > 0.05);Compared with the Shangjuxu group,low Dachangshu group the content of IL-10 in serum,the difference was statistically significant(P < 0.05),Zusanli group compared with no significant difference(P > 0.05).Conclusion:1.Electroacupuncture treatment of acute colitis,there is no significant difference in the efficacy of the four acupoints.2.Electro acupuncture treatment of chronic colitis for 3 days,there was no significant difference between the efficacy of four acupoints Electroacupuncture;10 days later,the curative effect is better than that of other acupoints at Tianshu.3.In the treatment of acute and chronic colitis,the levels of MPO,serum IL-6 and serum IL-10 in the colon tissue of rats with acute and chronic colitis were improved in different degrees.4.The mechanism of electroacupuncture at four acupoints on the treatment of acute and chronic colitis may be related to the regulation of MPO,serum IL-6 and serum IL-10 content in colon tissue.
