Effect Of Moxibustion On The Levels Of Serum Inflammatory Factor,?eta-defensin-2 And Beta-defensin-2 Gene In Colon Of Rats With Diarrhea Predominant Irritable Bowel Syndrome

1.Aim To observe and compare the level of serum interleukin(IL)-1?,IL-6,tumor crosis factor(TNF)-?,and IL-10,and the expression of ?-defensin-2(BD-2)protein and m RNA in colon tissue of diarrhea predominant irritable bowel syndrome(D-IBS)rats which was induced by the method of restraint stress plus senna leaf soak intragastric perfusion treated with moxibustion on Tianshu and Shangjuxu.To explore the partial mechanisms of treating D-IBS with moxibustion on Tianshu(ST25)and Shangjuxu(ST37).2.Methods 40 SD rats were randomly divided into 4 groups: blank control group,model group,medication group and moxibustion group(10 rats in each).Three groups of rats were treated with restraint stress plus senna leaf soak intragastric perfusion to establish D-IBS model except the blank control group.After modeling,the medication group was treated with trimebutine maleate intragastric perfusion with 50 mg/kg/d for 7 times.The moxibustion group was treated with mild moxibustion at each acupoint for 15 min/d for 7 days.Blank control group and model group are controlled only to eliminate other factors interference.The general state and diarrhea index of rats in different time points were observed and measured during the experiment.After anesthesia,abdominal aorta blood was drawn from rats in each group,and some colonic tissues were collected,and then killed the rats.Colonic sections were treated with HE staining,and the level of IL-1?,IL-6,TNF-? and IL-10 in serum of rats in each group was detected by ELISA kit.The expression level of ?-defensin-2 in colon tissue was detected by Wester blot.The expression level of ?-defensin-2 m RNA in colon tissue was detected by RT-PCR.Finally,the statistical softwares,such as Excel and SPSS,were used to compare the difference of measurement data,and the experimental results and conclusions are obtained.3.Results 1)General state of rats: The rats in blank control group had a normal diet,white and lustrous fur color,sensitive movement,very few bite and fight during the course of experiment.After modeling,the defecation of the other three groups increased,thinned,the color of the coat withered,and some rats were obviously thin and often bitten each other.After the treatment period of the medication group and moxibustion group,the defecation,coat color,diets were recovered.2)Changes of diarrhea index: After modeling,the diarrhea index of model group,medication group and moxibustion group was significant higher than that of the blank control group(all P < 0.05);there was no significant difference among the model group,medication group and moxibustion group(P > 0.05).After the treatment,the diarrhea index of the medication group and the moxibustion group were significantly lower than that of the model group(both P < 0.05).There was no significant difference among the medication group and moxibustion group(P > 0.05).3)Pathological sections of colon tissue were observed in 200×.No pathological changes and inflammatory infiltration were found in the control group.After modeling,there was a low degree of inflammatory reaction in the colonic mucosa,no obvious erosion and ulcer.Compared with the model group,after treated with trimebutine maleate and moxibustion on Tianshu and Shangjuxu,the colonic tissue and mucosal structure of the two treatment groups were intact,and no obvious neutrophil infiltration was observed in the two groups.4)Comparison of serum levels of IL-1?,IL-6,TNF-? and IL-10: The level of serum IL-1?,IL-6,TNF-? in the model group was significantly higher than that in the control group(all P < 0.05)and the level of IL-10 was decreased(P < 0.05).Compared with the model group,the level of IL-1?,IL-6,TNF-? in medication group moxibustion was decreased(all P < 0.05),the level of IL-10 was increased(all P < 0.05).In moxibustion group,the levels of IL-1? and IL-6 were similar to medication group(both P > 0.05).The level of serum TNF-? was higher than that in moxibustion group(P < 0.05),the level of serum IL-10 was lower than that in moxibustion group(P < 0.05).5)The comparison of colon tissue ?-defensin-2 and its m RNA in each group: The expression levels of colon tissue BD-2 and its m RNA in the model group were significantly higher than that in the blank control group(all P < 0.05).The levels of colon tissue BD-2 and its m RNA in medication group and moxibustion group were lower than that in blank control group(all P < 0.05).The expression levels of colon tissue BD-2 and its m RNA in the moxibution group were significantly lower than that in the medication group(all P < 0.05).4.Conclusions 1)Moxibustion on Tianshu and Shangjuxu could relieve the diarrhea symptom and low degree inflammation of colon in D-IBS rats,and the general state of rats was also recovered.2)Moxibustion can regulate the level of serum IL-1?,IL-6,TNF-?,IL-10 to make them normal.3)Moxibustion could decrease the expression levels of ?-defensin-2 protein and ?-defensin-2 m RNA in colon of D-IBS rats.
