| Objective: This paper Moringaceae Moringa plant Moringa seeds as the research object,on the various parts of type II diabetes hypoglycemic experiment,and isolated compounds,in order to provide some reference for the research and development of drugs reduced blood glucose.Methods: Firstly,different extraction solvents of Moringa seed get Moringa seed water extract and alcohol extract of Moringa seed parts and Moringa seed water extract alcohol parts.Then the model of type II diabetes was made in mice,and the mice with successful model making and good state were selected to carry out the hypoglycemic experiment.The use of one-way ANOVA and paired samples T test,get Moringa seed ethanol extract were the best hypoglycemic effect,and less influence on body weight of mice.After respectively with 30% ethanol,50% ethanol,70% ethanol,95% ethanol extract of Moringa seed extracts by different concentrations of ethanol,the type II diabetes hypoglycemic experiment,results showed that 30% ethanol extract hypoglycemic effect is the best,it is speculated that the 30% ethanol extract were containing more hypoglycemic chemical monomer component.Then,the high resolution liquid chromatography technology was used to analyze the chemical composition of 30% ethanol extract parts.,18 compounds and 14 main compounds were identified,and the vanillin content and the extraction process were optimized.Then the main monomer compounds with macroporous resin and semi preparative liquid phase separation technique in Moringa seed.Results: Type II diabetes hypoglycemic experiment showed that 30% ethanol extracts of Moringa seed hypoglycemic effect is the best.Reverse HPLC measurement of vanillin content of Moringa seed was 0.0320mg/g.The optimum extraction process of vanillin was as follows: the ratio of material to liquid was 1:14,the time of shock was 60 min,and the temperature was 55.Niazirin(1),niazimicin(2),1-O-phenyl-alpha-L-rhamnopyranoside(3),N,N ’-bis{4-[(alpha-l-rhamnosyloxy)benzyl]}thiourea.(4)were obtained by macroporous adsorption resin and semi preparative liquid phase separation.Through the analysis,it can be seen that the composition of the monomer is with figure 1 as the parent nucleus.Conclusion: In this paper,Moringa seed as the research object,carries on the study on hypoglycemic effect of different parts of Moringa seed found that the 30% ethanol extract had the best effect.Then,high resolution liquid chromatography-mass spectrometry(HPLC-MS)was used to analyze its compounds,and the content of vanillin was determined by reverse high performance liquid chromatography,and the extraction process was optimized.Four monomers were separated from them. |
