Expression And Clinical Significance Of ILF2 In Gastric Cancer

Objective: The aim of this research is to study the expression difference of ILF2 in gastric cancer tissues and corresponding adjacent normal gastric tissues at mRNA and protein levels,to analyze the relationship between ILF2 protein expression and clinical pathological characters of gastric cancer patients,to discuss the correlation between ILF2 protein expression and clinical prognosis of patients with gastric cancer,to assess the possibility of ILF2 protein as an independent prognostic indicator in gastric cancer patients.Methods: To detect the expression levels of ILF2 mRNA and protein,21 paired fresh gastric cancer tissue specimens and corresponding adjacent normal gastric tissue specimens,collected from Department of Gastrointestinal Surgery of the First Affiliated Hospital of Anhui Medical University,were respectively examined by quantitative Real Time-PCR(qRT-PCR)and Western Blotting(WB)assay.The expression difference of ILF2 mRNA and protein in two groups was analyzed by Paired T-test.In order to clarify the relationship between ILF2 protein expression levels and clinicopathological parameters in gastric cancer,immunohistochemistry(IHC)assay was performed in the paraffin-embedded sections of gastric cancer and corresponding adjacent normal gastric.The expression difference of ILF2 protein in two groups was analyzed by Wilcoxon matched-pairs signed-ranks test.In addition,using Kaplan-Meier survival curve to calculate the Overall survival time(OS)and Disease-free survival time(DFS)of patients with gastric cancer,and the significance of difference between two groups was analyzed by Log-Rank test.At last,the Cox ' s proportional hazard model was applied to analyze the prognostic factors of gastric cancer patients.Results: The expression levels of ILF2 mRNA and protein in gastric cancer tissues were relatively higher than that in corresponding adjacent normal gastric tissues(both P < 0.05).Besides,the results of Person correlation analysis revealed that the positive correlation was observed between ILF2 mRNA expression and ILF2 protein expression(r = 0.753,P < 0.001).According to the immunohistochemical results,it was founded that the positive expression rate of ILF2 protein in gastric cancer tissues was 61.67%(37/60),which was dramatically higher than 20%(12/60)of para-carcinoma gastric tissues,and the expression levels between two groups were greatly different(P < 0.001).In addition,the expression levels of ILF2 protein in gastric cancer tissues were related to the degree of differentiation(P < 0.001),depth of invasion(P = 0.046),lymph node metastasis(P = 0.009),and TNM staging(P < 0.001),but no significant association was discovered with sex(P = 0.986),age(P = 0.471),tumor location(P = 0.951),tumor size(P = 0.755),operation procedure(P = 0.0776),CA19-9(P = 0.464),CEA(P = 0.707),and CA125(P = 0.602).On the basis of Kaplan-Meier survival results,it showed that both OS and DFS of gastric cancer patients with ILF2 protein high expression were shorter than those in patients with ILF2 protein low expression,and Log-Rank test displayed that the differences between two groups were statistically significant(both P < 0.001).Cox ' s proportional hazard model clarified that ILF2 protein expression(RR = 3.464,P < 0.001),depth of invasion(RR = 3.669,P = 0.002),CA19-9(RR = 3.210,P = 3.210),CA125(RR = 2.391,P = 2.391)were the independent risk factors for DFS in patients with gastric cancer.Similarly,TNM staging(RR = 3.462,P = 0.005),ILF2 protein expression(RR=2.996,P=0.022),depth of invasion(RR = 3.849,P = 0.007),CA19-9(RR = 2.911,P = 0.004),CA125(RR = 3.632,P = 0.001)were all the independent prognostic indicators for OS of patients with gastric cancer.Conclusion:(1)The abnormal high expression of ILF2 in gastric cancer tissue may be closely related to the occurrence of gastric cancer.(2)The expression of ILF2 protein in gastric cancer tissues is associated with histological differentiation,depth of invasion,lymph node metastasis,and TNM staging.Moreover,the higher expression of ILF2 protein is,the relatively more malignant degree of gastric cancer is.Consequently,it is suggested that ILF2 protein may highly associate to the progression of gastric cancer.(3)The expression of ILF2 protein is related to the clinical outcomes and DFS in patients with gastric cancer,and it can be concluded that ILF2 protein may act as an important indicator to predict the prognosis and monitor the recurrence of gastric cancer.
