Objective To investigate the expression of SRF protein in gastric cancer tissue and thetissue adjacent to carcinoma. We analyzed the correlations between SRF protein expressionand clinical pathologic materials,further analyzed the correlations between SRF proteinexpression and5-year survival rate in patients with gastric cancer after surgical treatment.Theresearch is aimed to assess whether SRF could be used as a biological marker of gastriccancer and assist in the early diagnosis of gastric cancer and to evaluate prognosis of gastriccancer.Methods The expression of SRF protein in the96cases of gastric cancer(GC) tissues and96cases of adjacent noncancerous tissues (ANT) was examined byStreptavidin-Peroxidase(SP) immunohistochemistry.And the relationships was evaluatedbetween SRF expression and clinicopathological characteristics.In addition, these patientswere followed up to investigate the relationships between SRF expression and the survivaltime.Results The positive rate of SRF was83.3%in gastric cancer tissues, significantly higherthan adjacent noncancerous tissues (31.3%, P<0.001). There was a close relationshipsbetween SRF expression and TNMl stage (P=0.001) and the lymph nodemetastasis(P=0.026),but not with gender (P=0.685), age(P=0.648),and the degree ofhistological differentiation(P=0.886),T stage(P=0.143)The Univariate analysis demonstratedthat the5-year survival time of patients was significantly correlated with age, LN metastasis,TNM stage and the intensity of SRF expression (P<0.05), with on obvious relation withgender, T stage and differentiation (P>0.05). In order to further analyze that the expression of SRF is of significance in GC, the Multivariate analysis indicated that LN metastasis and SRFexpression were two independent prognostic factors to poor prognosis. The intensity of SRFprotein expression is closely related to survival time.The5-years survival rate were68.7%,52.6%,47.4%and21.7%respectively,P=0.027.The difference was statistically significant.Conclusion SRF might be regarded as one of the useful molecular markers to diagnoseand predict the prognosis of gastric cancer patients.
