ObjectiveFrom the perspective of the world,the incidence of gastric cancer ranked fourth in malignant tumors;for our China,the incidence and mortality of gastric malignant tumor is higher,ranked second in malignant tumors,the number of deaths due to the malignancy disease ranked first in the digestive system malignancy.Radical surgery is the only possible cure means for early gastric cancer currently,However,due to the early symptoms of gastric cancer is not typical,patients often have no specific symptoms,coupled with the popularity of gastric cancer screening is not high in our country,more than half of the patients have been in the advanced stages when they go to hospital,and lost the chance of radical surgery.Surgical treatment combined with radiotherapy and chemotherapy has made some progress in the treatment of advanced gastric cancer,but the patient's prognosis and quality of life is still an urgent need to improve.Therefore,we need to research the occurrence and development mechanism of gastric cancer deeper,to find more sensitive and specific biological markers which contribute to early detection and intervention in gastric cancer,predict the prognosis of patients and make more effective anti-tumor drugs,which is an important part of gastric cancer prevention and cure work.Brain-selective protein kinase-2(BRSK2)is a member of the AMP-activated protein kinase(AMPK)family,early research of BRSK2 focused on its functionrelates to the nervous system.With the deepening of research,researchers have found that BRSK2 can regulate cell cycle and affect cell proliferation and apoptosis,which plays an important role in the tumorgenesis and progression of pancreatic cancer and other tumors.However,the expression of BRSK2 in gastric cancer and its correlations with the prognosis of gastric cancer patients,No related reports have been seen and need to be clarified.Therefore,we studied the BRSK2 expression in human gastric cancer tissue and the corresponding normal gastric mucosa tissue and its relationship with the clinical and pathological parameters of patients,We also combined with the patient's survival data analyzed the relationship between BRSK2 expression and the prognosis of patients with gastric cancer.MethodsOf 121 cases of gastric cancer tissues and corresponding normal tissues,the expression of BRSK2 protein was detected by immunohistochemistry.We analyze the relationship between BRSK2 and clinical pathological parameters in patients with gastric cancer.The relationship between BRSK2 expression and its correlations with the survival of gastric cancer patients was further analyzed by patients' follow-up data(Kaplan-Meier method and Log-rank test);Finally,Cox regression model was established to evaluate the correlation between BRSK2 and clinical pathological parameters and prognosis of gastric cancer patients.Results1.BRSK2 decreased expression in 62 of 121 cases of gastric cancer(51.24%),which was significantly higher than adjacent noncancerous epithelialtissues(c2=53.06,P<0.001).2.BRSK2 decreased expression was significantly correlated with Depth ofinvasion/T classification(P<0.001),Histological differentiation(P=0.006),TNM stage(P<0.001),and Lymph node metastasis(P=0.001).However,it wasnot associated with Age,Gender,Location,Diameter,Surgical,Carcino-embryonic antigen(CEA)and Carbohydrate antigen 19-9(CA19-9).3.Kaplan–Meier survival analysis showed that the overall survival(OS)timeand disease-free survival(DFS)time in patients with BRSK2-negativeexpression were shorter.4.Univariate Cox regression analysis showed that cancer tissue differentiation,tumor invasion depth,lymph node metastasis,TNM stage,CEA,CA199 andBRSK2 affected the prognosis of gastric cancer patients;Multivariate Coxregression analysis showed that the age of patients,BRSK2,TNM tumorsand CEA were independent prognostic factors for gastric cancer patients.Conclusion1.BRSK2 protein decreased expression in gastric cancer tissues which showedsignificant difference with normal gastric mucosa2.the reduction of BRSK2 expression level is involved in the occurrence ofgastric malignancy and tumor progression,and relates to the poor prognosis ofpatients with gastric cancer.3.BRSK2 can be used as a molecular markers for predicting the prognosis ofgastric cancer patients and a potential target for tumor therapy.
