Colorectal cancer is the third leading incidence of malignant tumor in the world. The majority(about 80%)of colorectal cancers arise sporadically in a time-dependent manner through colonic polyp formation.Colonoscopy is the gold standard for the diagnosis of polyps. But its high cost and troublesome make it difficult to be accepted which may cause a higher incidence of cancer. Therefore,it is necessary to explore a new and effective screening method to amplify the tense pattern. With the latest development of generation sequencing technology,it has found that intestinal microbiome is closely related to human health and disease. It has been verified the change of the structure and diversity of intestinal flora in patients with colorectal adenoma suggesting the potential of microbiome as a biomarker for polyps. In this subjects high-throughput sequencing technology of 16S rDNA and ITS2 combining R analysis has been used to explore the changes of microbial community and diversity in patients with polyps. The result shows an obvious change of intestinal microbial community.Drease of diversity has happened both in bacteria and fungi in patients with polyps. Also, some differential flora has been found in different level of classification. Then the differential flora in geneus or species level was obtained by the means of Lefse analysis.By the means of Lefse analysis,it comes out that a higher abundance of pathogenic bacteria(such as Bacteroides,Parabacteroides,Fusobacterium genus)and opportunistic fungi(such as Candida and Aspergillus)in polyp patients. In addition, the ROC curve analysis shows a pretty good accuracy of thses flora to identify the patients, which is up to above 0.7.In summary, this study investigats the microbial community structure of polyp patients and elucidate the characteristics of patients with pathogenic or opportunistic bacteria and fungi. What's more, this subjects has established innovative intestinal microorganisms as polyp screening biomarkers.
