Objective: Integrin alpha 2(ITGA2) is an important member of the i ntegrin, which at the 807 C/T site have a gene polymorphism(rs1126643). Studies have shown that this locus gene polymorphism have some associated with many diseases, such as myocardial infarction, cerebral infarction ect. And think that cerebral infarction has a certain relationship with lipid metabolism. for this purpose, we do some an-alysis research on polymorphis-m of the ITGA2 gene- 807- c/T of the ha-n population in north guangxi in guangxi region by using polymerase chain reaction- restriction fragment length polymorphism(PCR- RFLP) technique, at the gene level to discus-s ITG-A2 gene polymorphism and the relationship betw een DN, which may for the early diagnosis and treatment of diabetic nephropathy provide new train of thought at the genetic level. Methods: Selection 206 cases type 2 diabetes of han population in north guangxi region,who according to the diagnostic criteria of diabetic nephropathy were divided into diabetic nephropath-y(DN) group and non diabetic nephropathy diabetes(T2DM) two groups, and choice 90 cases of healthy check-up at the same term as healthy controls(NC) groups. The serum lipid and apolipoprotein levels were measured byroutine method. And the genotypes and allele frequencies distribution were assessed by Polymerase chain reaction-restriction fragment length Polymorphi-sm(PCR-RFLP).Results: ITGA2 gene- 807 gene of han people in North region of guangxi have three kinds of genotype CC、CT and TT, The genotype frequencies in the NC group were 72.2%, 21.1% and 6.7% respectively, All ele frequency is C = 88.7% and T = 11.3%; In T2 DM group were 59.2%, 32.7%, 8.2%, and allele frequency is C = 75.5%, T = 24.5%; In the DN group were 49.1%, 42.6%, 8.3%, and allele frequency is C = 70.4%, T = 29.6%.Compared with control group, the DN group blood TC, TG, LDL-C values were elevated(P < 0.05),DN group ITGA2 gene- 807- C/T genotype distribution and allele frequency were statistically significant(P < 0.05);CT + TT T allele frequency and genotype was significantly higher than that of control group(all P < 0.05); The individual who carry the T allele have the 1.72 times risk of DN in who carry C allele(OR = 1.72, 95% CI = 1.72 ~ 2.517). The T2 DM group compared with the control group and the DN group, ITGA2 gene- 807- C/T genotype distribution and allele frequency differences had no statistical significance(P > 0.05). DN patients of TC, TG, LDL- C lipid levels than healthy controls were increased obviously, and blood lipids TC levels are homozygous CC genotype patients carry T genotype(CT + TT type) of DN patients significantly decreased, combined with logistic regression analysis, TC、LDL-C are important risk factors influence the pathogenesis of DN.Conclusions: The polymorphism of ITGA2-807C/T gene is associated with the development of DN in Han nationality in northern part of Guangxi. the T allele may be a genetic susceptibility genes which promote the occurrence of DN. T allele can promote the level of blood lipids TC high expression and increase the risk of DN.
