Research background:Biophoton radiation is the essence of life system,which carries the whole information of life system.The study shows that biophoton radiation detection can get the overall information of the measured biological effect of the samples caused by internal and environmental impact,can provide a comprehensive index about biological characteristics,so that the photon emission detection is a quantitative detection method with the theory of traditional Chinese medicine on the whole.Research purpose:This paper based on "biological photon coherence" theory,using bio photon radiation detection technology by Scenedesmus ultra weak chemiluminescence detection and optimization of experimental conditions,to study the feasibility of Scenedesmus as biological indicator.Finally,as a biological indicator,a method system which can not only accord with the holistic view of Chinese medicine,but also accord with the modern cognition,is established.Research method:In order to Scenedesmus as a biological indicator,using bio photon radiation detection technology,measurement with Scenedesmus biophoton emission in decoction,and then use the double exponential decay model e model to analyze the experimental results and the parameters of Gu ",with the combination of curve fitting,linear regression and ROC curve analysis and data processing method,identification of cold of traditional Chinese medicine and drugs.Research content and results:1、 based on the BG11 medium,contrast analysis,biophoton emission of Rehmanniae Decoction and Scenedesmus solution can be found,medium and Rehmannia Decoction has no biological photon radiation,which precludes the two experimental process self luminous effect on Scenedesmus biophoton emission measurement.Next,add dried Rehmannia Decoction after detection of Scenedesmus delayed luminescence,results showed that Radix Rehmanniae Decoction will biophoton emission behavior of Scenedesmus were significantly influenced by the Scenedesmus,spontaneous emission intensity and effect of delayed luminescence trend of initial strength increased first and then decreased,and in a certain range.The spontaneous emission intensity of Scenedesmus and Rehmannia decoction volume showed good linear relationship.This shows that the use of Scenedesmus as indicator has high feasibility.2、 The experiment was carried out to optimize the use of the indicator,the dosing volume and the time interval of the delayed luminescence measurement:(1)used as a biological indicator of Scenedesmus method has two kinds,one is the sample to be tested directly into the algae fluid,biophoton radiation measurement of algae liquid;the other is the sample to be tested into the algal cells were measured.Therefore,to salina algae cells and bio photon emission in this paper were measured,and experimental results show that the luminescence intensity is higher than the algae algae liquid,in the traditional Chinese medicine decoction,adding,biophotons algae liquid radiation has significant changes in algal cells showed no change.Therefore,the suitable method for the use of Scenedesmus as indicator is that the sample to be tested is directly added into the liquid to measure the biological radiation of the algae.(2)experimental design of the 6 dosing volume,respectively 50μl,100μl,150μl,200μl,250μl,300μl,the use of Gu model parameters on the delayed luminescence of experimental data analysis,y= Kt+b linear equation can be obtained with different dosing volume of Scenedesmus the delayed luminescence intensity and time average,because the will to identify Chinese medicine cold and heat resistance,the slope of the K linear equations so that the linear equation fitting degree should be better.The final results show that the linear equation fitting degree is the highest when the dosage is 100μl,which is 96.36%.(3)different time intervals were used to measure the delayed luminescence,respectively 0min,5min,10 min,15min.The experimental results show that the different time delay has no effect on Scenedesmus,luminescence measurements.However,when the time interval is 10 min when the measured data is the most stable,that is to say,the optimal delay time interval Scenedesmus chemiluminescence detection for 10 min.3、 The total amount of the traditional Chinese medicine decoction was measured by using Scenedesmus sp.as indicator.A total of 80 traditional Chinese medicines were measured.Firstly,the delayed luminescence curve were analyzed by e double exponential function model,we can get five parameters,namely yo,A1,A2,T1,T2,showed that T2 has as a traditional Chinese medicine cold heatnature discriminant index the possibility of further use of ROC curve analysis,can be found T2 discriminant effect on heat and cold traditional Chinese medicine however,the accuracy is relatively low.Then use the "Gu parameters" mathematical model of delayed luminescence curve fitting analysis,obtained three parameters Iw,Io,S,and the establishment of the time t and Scenedesmus delay linear relationship between the average intensity of luminescence of Y,the linear equation of the two: y=Kt+b.Through the analysis,we found that the K of the Chinese herbal medicine was significantly greater than that of the traditional Chinese medicine,p<0.001.ROC curve analysis results slope K shows: AUC=0.868,p<0.001,K have the effect on the slope that distinguish heat and cold traditional Chinese medicine,and higher accuracy,the threshold is 4.43,the sensitivity was 77.8%,specificity was 81.8%,Youden index was 0.596.To sum up,light delay based on Scenedesmus,we get a reliable parameter to distinguish indirect cold hot natures,namely the linear equation of the slope of y=Kt+b.
