| Objective:To explore the effects and molecular mechanism of electroacupuncture(EA)at shu and mu points of stomach on the gastric motility of functional dyspepsia(FD)rats and provide experimental basis and new ideas for the application of Shujun acupoint.Method:Male clean sprague-dawley(SD)rats were randomly divided into five groups:blank group,model group,Zhongwan(CV12)+Weishu(BL21)group,Weishu(BL21)group and Zhongwan(CV12)group.The FD model was established by the combination of moderate tail irritation and irregular feeding.In addition to blank group,all rats were fixed with a self-made fixator,and the Zhongwan Weishu group,Weishu group and Zhongwan group were treated with electroacupuncture under the sober state.Gastrointestinal pressure transducer was connected to LabChart software system to monitor gastric motility;colorimetric method was used to detect the Glu expression of neurotransmitters in the dorsal motor nucleus of the vagus;the expression of NR1 subunit of NMDA receptor in rat DMV nuclei was detected by immunohistochemistry;the expression of NR2(A-D)protein in DMV of Rats Nucleus was detected by Western blot;ELISA was used to detect the expression of NO and P in the serum of rats.Results:1.FD model evaluationThe model rats were observed to be out of control,irritable and aggressive,bite each other when they were slightly irritated.Compared with blank group,the model rats had coarse hair,loose stool and reduced the amount of food and water consumed.In comparison with blank group rats,the contractile amplitude of gastric motility was decreased in model rats,but,no obvious organic lesion changed in the anatomical between model group and control group.2.Effect of electroacupuncture on gastric antral motility in ratsCompared with the blank group,the amplitude of gastric antrum movement in the model group decreased(P<0.05);Compared with the blank group,the amplitude of gastric antral motility was increased in the Zhongwan+Weishu group,Weishu group and Zhongwan group(P<0.05);and the gastric antral motility amplitude in the Zhongwan Weishu group was higher than that in the Zhongwan group and Weishu group(P<0.05).there was no significant change in the frequency of antral movement in each group(P>0.05).3.Effect of electroacupuncture on Glu expression of neurotransmitters in DMV nuclei of ratsCompared with the blank group,the expression of neurotransmitter Glu decreased in the model group(P<0.05);and the expression of Glu was increased in the Zhongwan group,the Weishu group and the Zhongwan+Weishu group in omparison with model(P<0.05).4.Effect of electroacupuncture on NR1 expression of DMV Nucleus in RatsThe expression of NR1 proteins was decreased in model group in comparison with blank group(P<0.05);the expression of NR1 proteins in model group was lower than Zhongwan group and Weishu group and Zhongwan+Weishu group(P<0.05).5.Effect of electroacupuncture on the expression of NR2(A-D)in DMV Nucleus in RatsCompared with the model group,the NR2 A in the Zhongwan+Weishu group and the Zhongwan group was significantly increased(P<0.05);compared with the model group,the NR2 B was significantly increased in the Zhongwan+Weishu group and the Weishu group(P<0.01);compared with the model group,the NR2 B increased significantly in the Zhongwan+Weishu group and the Weishu group.Compared with the model group,the changes of NR2 D in Zhongwan+Weishu group,Weishu group and Zhongwan group were not statistically significant(P>0.05).6.Effect of electroacupuncture on the expression of serum NO/P in ratsCompared with the blank group,the expression of serum NO in the model groupincreased(P<0.05);Compared with the model group,the expression of NO in serum decreased in the Zhongwan+Weishu group,Weishu group and Zhongwan group(P<0.05);Compared with Weishu group and Zhongwan group,the expression of NO in serum was decreased in Zhongwan+Weishu group(P<0.05);Compared with the blank group,the expression of serum substance P in the model group decreased(P<0.05);Compared with the model group,the serum substance P expression in the Zhongwan+Weishu group,Weishu group,Zhongwan group increased(P<0.05).Conclusion:1.DMV is an important central target for regulating stomach movement by electroacupuncture AT shu and mu points of stomach,and its neurotransmitter Glu and NMDA receptors may play an key role.2.Electroacupuncture on shu and mu points of stomach could increase gastric movement of FD rats,and the mechanism may be involved in the changes of gastrointestinal hormone NO and P substances.3.Electroacupuncture at shu and mu points of stomach could regulate the gastric movement of FD rats,and there is a synergistic effect of combination of point Zhongwan and Weishu.The molecular mechanism was that the regulation of Glu-NMDA receptor signal transduction in the DMV region to regulate the expression of gastrointestinal hormone NO and P substances in serum. |
