Objective 1.To study the effect of mangiferin on SHR blood pressure;2.Observe the changes of mangiferin on related vascular morphology;3.To investigate the effect of mangiferin on the Mc P-1/CCR2 signaling pathway in thoracic aorta;4.Preliminary study on the efficacy of mangiferin to suppress liver Yang;Methods Male spontaneously hypertensive rats(SHR)48,randomly divided into 6 groups,each group of eight: blank control group,model group(SHR),benzene the captopril group(10 mg/kg.d),mangiferin low dose group(25 mg/kg.d),middle dose group(50 mg/kg.d),high dose group(100 mg/kg.d),in addition to choose with weeks of male WKY rats as normal control group.Take blood pressure in each group of rats every two weeks;HE staining to observe the thoracic aorta,the carotid artery and superior mesenteric artery pathological changes;Immunohistochemical method to detect the thoracic aorta,CCR2 and MCP-1 protein expression changes;Western blot method to detect thoracic aorta,the protein expression level of CCR2 and MCP-1;Real-Time PCR detection of thoracic aorta MCP-1,CCR2 gene expression level;Observation the biological characteristics of rats;Adopt the method of HPLC for determining the content of plasma epinephrine in spontaneously hypertensive rats.Results 1.blood pressure of rats in each group: compared with groupWKY,blood pressure in group SHR was significantly higher than that in group WKY(P <0.01).Compared with group SHR,the blood pressure of mangiferin dose group and benazepril group decreased significantly,P <0.01;2.The pathological changes of the thoracic aorta,the carotid artery and the superior mesenteric artery were observed by HE staining.Compared with the WKY group,the fibrous tissue of the SHR group of the thoracic aorta was hypertrophy,the endothelial cells fell off,the middle membrane was thickened,the inflammatory cells increased and the endothelial edge was rough.Compared with the group SHR,the mangiferin dose group and the benazepril group were obviously reduced.Less,the edge of the endothelium improved,the infiltration of fibrous tissue improved significantly,the endothelium was also smoother,and there was no significant change in the normal group.Compared with group WKY,the content of MCP-1 and CCR2 protein in group SHR was significantly higher than that in group WKY(P<0.01).Compared with group SHR,the content of MCP-1 and CCR2 proteins in the benazepril group was significantly lower than that in SHR group(P),and there were significant differences in the dose group and high dose group in the mangiferin;3.The results of Western blot assay showed that compared with the WKY group,there was a significant difference in the expression of MCP-1 and CCR2 protein in the group SHR(P <0.05).Compared with the SHR group,the MCP-1 and CCR2 contents of the thoracic aorta in each group were reduced in varying degrees;4.Real-Time PCR assay showed that the expression level of MCP-1 and CCR2 in the thoracic aorta of the treatment group was significantly increased(P <0.05,P <0.01);5.Groups of rats macroscopic results show: WKY group weight have arapid increase before 3 weeks,SHR group gain the slower better than treatment group,SHR group: easy to anger,exophthalmos,color slants yellow,defecate is dry,a dry moss,treatment group: depilate symptoms,defecate become soft,water slippery moss;HPLC method to detect the plasma epinephrine(A)content: compared with WKY group,significantly increased the content of adrenaline in SHR group,benzene the captopril group and mangiferin dose group compared with model group,no significant change in the adrenaline levels,but the adrenaline levels fell;Conclusion 1.Mangiferin can reduce the blood pressure of SHR,which is equivalent to benazepril;2.Mangiferin could improve the inflammatory injury of SHR thoracic aorta,carotid artery and superior mesenteric artery;3.Mangiferin significantly decreased the expression of MCP-1 and CCR2 in SHR thoracic aorta;4.Mangiferin could inhibit the expression of MCP-1 and CCR2 m RNA in SHR thoracic aorta;5.Mangiferin could reduce the macroscopic symptoms of SHR and decrease the level of epinephrine in plasma;...
